摘 要:WRKY是一类与植物生物胁迫反应相关的转录因子,是植物中特有的超基因家族。为了挖掘百合抗蚜虫胁迫相关的WRKY基因,本研究利用棉蚜胁迫的百合转录组数据,对WRKY进行筛选及保守结构域分析,对鉴定出的WRKY转录因子进行生物信息学分析。结果表明:百合中筛选的29个WRKY基因的蛋白为99~675个氨基酸,理论等电点的范围为5.06~11.19。在LhWRKY基因编码的所有蛋白中,其中LhWRKY5和LhWRKY26为稳定蛋白,其余LhWRKY属于不稳定蛋白。所有LhWRKY蛋白均定位于细胞核,属于亲水性蛋白。除LhWRKY2、LhWRKY3、LhWRKY5、LhWRKY20四个蛋白外,均具有丝氨酸(Ser)、苏氨酸(Thr)和酪氨酸(Tyr)磷酸化位点。LhWRKY包含三类WRKY转录因子,部分蛋白发生变异。系统进化树发现,有3个WRKY基因与已知抗生物胁迫基因同源性较高,推测其参与百合抗蚜胁迫反应。研究结果可以通过筛选的WRKY基因,进行进一步的功能验证,为植物抗生物胁迫尤其是抗蚜育种提供分子基础。WRKY transcription factor is related to plant biotic stress,which is a unique supergene family in plants.In order to explore the WRKY genes of lily related to plant biotic stress,based on the transcriptome data of lily under Aphis gossypi stress,WRKY was screened,the conserved domain analysis was performed and applied bioinformation analysis.The results showed that the 29 WRKY genes in the lily were 99~675 amino acids and the theoretical isoelectric point ranged from 5.06 to 11.19.Of all the proteins encoded by LhWRKY gene,only Lh-WRKY5 and LhWRKY26 were stable protein,and the rest were all unstable proteins.All proteins were located in the nucleus and belonged to hydrophilic protein.Except that LhWRKY2,LhWRKY3,LhWRKY5 and LhWRKY20,all other amino acids had Ser,Thr and Tyr phosphorylation sites.LhWRKY contained three types of WRKY transcription factors and some of proteins mutate.Phylogenetic trees revealed that 3 WRKY genes shared high homology with known biotic stress genes,and it was speculated that they participated in the aphid resistance of lily.The results of the study could be further verified by screening WRKY genes,which would provide a molecular basis for plant resistant breeding especial for aphid resistant breeding.
