摘 要:水稻油脂组分含量是影响水稻食味品质的重要因素之一。开发水稻油脂组分含量主效基因qPAL6的分型检测方法并开展分子标记辅助选择,是改良水稻油脂组分含量及培育优良食味品质品种的有效途径。本研究利用qPAL6位点的序列差异,开发了qPAL6基因的功能性InDel(Insertion/Deletion)标记PAL6M2;同时基于PARMS技术(Penta-primer amplification refractory mutation system,五引物扩增受阻突变体),开发了qPAL6基因内的荧光分子标记PM-PAL6。利用分子标记PAL6M2和PM-PAL6对96份水稻材料进行分析,结果表明,2个分子标记均可用于水稻qPAL6位点基因型检测。该研究为水稻油脂组分含量分子标记辅助育种提供了实用可靠的基因分型技术。The content of oil components is one of the important factors affecting the eating quality in rice.To im-prove the content of oil components and breed varieties with good eating quality,it is an effective way to develop a genotyping method for the major gene qPAL6 of oil component content and carry out molecular marker assisted se-lection.In this study,the functional InDel(insertion/deletion)marker PAL6M2 of qPAL6 gene was developed ba-sed on the sequence difference of qPA L6 locus,and the fluorescent molecular marker PM-PAL6 within qPAL6 gene was developed based on PARMS(Penta primer amplification refractory mutation system).PAL6M2 and PM-PAL6 were used to detect the genotypes of 96 rice lines.The results showed that there were genotypic differences betw-een the lines,and the two molecular markers could be used to detect the genotypes of qPA L6 locus in rice.This provides a practical and reliable genotyping technique for molecular marker assisted selection of oil content in rice.
