Apelin-13损伤大鼠新物体识别记忆再巩固及其cAMP-乳酸通路机制研究

摘　　要：目的评价侧脑室注射Apelin-13对大鼠新物体识别(NOR)记忆再巩固的影响,并探讨cAMP-乳酸通路是否参与Apelin-13介导的效应。方法行为学程序包括三个阶段:训练(第1天)、再激活(第2天)与检测(第3天)。在检测阶段记录辨别指数、对两物体总探究时间及穿越地板格子数,分别评价记忆能力、探究动机及自发活动能力;用ELISA法检测cAMP与乳酸水平。结果再激活后立即注射Apelin-13损伤NOR记忆能力(P<0.05);再激活后6 h或者无再激活条件下注射Apelin-13均不影响NOR记忆能力。此外,Apelin-13不影响探究动机与自发活动能力。Apelin-13下调海马组织cAMP(P<0.05)与乳酸水平(P<0.05);cAMP类似物8-Br-cAMP阻断Apelin-13引起的乳酸下调(P<0.01);8-Br-cAMP与L-乳酸均能挽救Apelin-13对NOR记忆再巩固的损伤作用(P<0.01)。结论Apelin-13通过下调cAMP/乳酸通路损伤NOR记忆再巩固。Objective To assess the effect of intracerebroventricular injection of Apelin-13 on novel object recognition(NOR)memory reconsolidation in rats,and to explore whether the cAMP-lactate pathway is involved in Apelin-13-mediated effect.Methods The Behavior procedure included three phases:training(day 1),reactivation(day 2)and testing(day 3).During the testing phase,the discrimination index,the total time spent exploring the two objects and the number of floor grids traversed were recorded to assess memory,motivation and spontaneous activity respectively;cAMP and lactate levels were measured by ELISA.Results Apelin-13 infusion immediately after reactivation impaired NOR memory performance(P<0.05).Apelin-13 infusion 6 h after reactivation or without reactivation did not affect NOR memory performance.Additionally,Apelin-13 had no effects on exploring motivation and locomotor activity.Apelin-13 downregulated the levels of cAMP(P<0.05)and lactate(P<0.05)in the hippocampus.A cyclic AMP analog8-Br-cAMP blocked Apelin-13-induced downregulation of lactate(P<0.01).Both 8-Br-cAMP and L-lactate rescued apelin-13-induced impairment of NOR memory reconsolidation(P<0.01).Conclusion Apelin-13 impairs NOR memory reconsolidation through downregulation of cAMP/lactate pathway.
