摘 要:目的从薏苡仁Coicis Semen中分离纯化多糖,分析其单糖组成并通过血清代谢组学探讨薏苡仁多糖对脾气虚证的调节作用。方法石油醚脱脂、热水抽提、乙醇沉淀、Sevage法除蛋白得到薏苡仁多糖(Coicis Semen polysaccharide,CP),经过DEAE-52纤维素离子交换柱、Sephadex G-75柱进行分离纯化,得到水洗脱多糖CP-W,气相色谱分析单糖组成;通过饮食失节(高脂低蛋白饲料)与劳累过度相结合的方法建立大鼠脾气虚证疾病模型,对获得的血清进行HPLC-TOF/MS分析,以期从代谢组学角度阐明薏苡仁多糖对其脾气虚证的调节作用。结果从薏苡仁中分离纯化得到CP-W多糖,其总碳水化合物含量为97.32%,单糖组成分析表明CP-W多糖由L-鼠李糖、L-阿拉伯糖、D-木糖、D-葡萄糖、D-半乳糖组成,单糖组成比例为0.4∶0.3∶1.1∶97.7∶0.5。与对照组相比,模型组在给予CP-W之后,在血清中发现12种差异代谢物,主要与脂质代谢、能量代谢和激素代谢相关。结论薏苡仁多糖能改善脾气虚证模型的生理特征,血清代谢组学结果表明薏苡仁多糖能够调节脾气虚证大鼠紊乱的脂质代谢、能量代谢、激素代谢等异常。Objective To isolate and purify polysaccharide from Coix lacryma,analyze its monosaccharide composition,and explore the regulatory effect of C.lacryma polysaccharide on spleen qi deficiency syndrome through serum metabolomics.Methods Coicis Semen polysaccharide(CP)was obtained by degreasing with petroleum ether,hot water extraction,ethanol precipitation and protein removal by Sevage method.After separation and purification by DEAE-52 cellulose ion exchange column and sephadexg-75column,the water washed polysaccharide CP-W was obtained,and the monosaccharide composition was analyzed by gas chromatography(GC);The disease model of spleen qi deficiency syndrome in rats was established by the combination of diet disorder(high-fat and low protein diet)and overwork.The obtained serum was analyzed by the HPLC-TOF/MS,in order to clarify the regulatory effect of CP on spleen qi deficiency syndrome from the perspective of metabolomics.Results CP-W polysaccharide was isolated and purified from Coicis Semen,and its total carbohydrate content was 97.32%,monosaccharide composition analysis showed that CP-W polysaccharide was composed of L-rhamnose,L-arabinose,D-xylose,D-glucose and D-galactose,The monosaccharide composition ratio was 0.4∶0.3∶1.1∶97.7∶0.5.Compared with the control group,12 different metabolites were found in the serum of the model group after CP-W administration,mainly related to lipid metabolism,energy metabolism and hormone metabolism.Conclusion CP could improve the physiological characteristics of spleen qi deficiency syndrome model.The results of serum metabolomics show that CP can regulate the disorder of lipid metabolism,energy metabolism,hormone metabolism and other abnormalities in rats with spleen qi deficiency syndrome.
