九味清脂膏改善非酒精性脂肪肝炎小鼠肝脏炎症模型疗效观察

摘　　要：目的 观察九味清脂膏对非酒精性脂肪肝炎(NASH)小鼠的肝脏炎症的改善作用。方法 将30只C57BL/6小鼠随机分为正常喂养组、模型对照组、多烯磷脂酰胆碱组(150 mg·kg^(-1))、九味清脂膏低剂量组(1.56 mg·kg^(-1))、九味清脂膏高剂量组(6.24 mg·kg^(-1)),每组6只。除正常喂养组外,其余小鼠均通过喂养高脂饲料8周制备NASH小鼠模型,然后分别予多烯磷脂酰胆碱、九味清脂膏灌胃给药,持续4周。检测肝脏湿重、体质量、血清丙氨酸氨基转移酶(ALT)、血清天门冬氨酸氨基转移酶(AST)、血清甘油三酯(TG)、血清总胆固醇(TC);HE染色观察肝脏病理特征以及非酒精性脂肪性肝病活动度积分(NAS)。结果 与模型对照组对比,九味清脂膏低剂量组和高剂量组小鼠体质量均有显著减少(P<0.05);九味清脂膏低、高剂量两组小鼠的ALT、AST、TC、TG水平均有明显下降(P<0.05)。病理提示九味清脂膏高剂量组可明显改善肝细胞的脂肪变性及炎症状态,且优于多烯磷脂酰胆碱组。结论 九味清脂膏对高脂饮食诱导的NASH小鼠的肝脏炎症有的改善作用。Objective To observe the effect of Jiuwei Qingzhi Ointment(九味清脂膏)on liver inflammation in nonalcoholic steatohepatitis(NASH)mice.Methods Thirty C57BL/6 mice were randomly divided into normal feeding group,model control group,polyene phosphatidylcholine group(150 mg·kg^(-1)),low-dose Jiuwei Qingzhi Ointment group(1.56 mg·kg^(-1))and high-dose Jiuwei Qingzhi Ointment group(6.24 mg·kg^(-1))with 6 mice in each group.Except the normal feeding group,other mice were fed with high-fat diet for 8周eeks to prepare NASH mouse models,and then were given polyene phosphatidylcholine and Jiuwei Qingzhi Ointment by gavage for 4 weeks.The liver wet weight,body weight,serum alanine aminotransferase(ALT),serum aspartate aminotransferase(AST),serum triglyceride(TG)and serum total cholesterol(TC)were measured.HE staining was used to observe the pathological characteristics of liver and the activity score(NAS)of nonalcoholic fatty liver disease.Results Compared with the model control group,the body mass of mice in the Jiuwei Qingzhi Ointment low-dose and high-dose groups decreased significantly(P<0.05).The levels of ALT,AST,TC and TG in mice of Jiuwei Qingzhi Ointment low-dose and high-dose groups decreased significantly(P<0.05).Pathology showed that the Jiuwei Qingzhi Ointment high-dose group could significantly improve the steatosis and inflammation of liver cells,and was superior to the polyene phosphatidylcholine group.ConclusionJiuwei Qingzhi Ointment has therapeutic effect on NASH mice induced by high-fat diet.
