摘  要:[目的]探讨TCF3在胃癌组织中的表达水平及其与临床预后的相关性。[方法]通过GEPIA、ULCAN数据库分析TCF3在胃癌组织中的差异性表达、临床病理资料以及与错配修复基因之间的关系,并利用Kaplan-Meier Plotter数据库绘制TCF3与胃癌患者生存预后曲线。根据Spearman相关系数寻找胃癌中和TCF3作用相关的基因,另外对TCF3及其相关基因进行G0/KEGG富集分析,找出其中最具差异性的生物学注释,最后使用String数据库搭建TCF3蛋白质和上下游蛋白的调控互作网络。[结果]胃癌组织中TCF3表达水平明显高于正常胃组织(P<0.05),而且TCF3与错配修复基因MLH1、MSH2、MSH6、PMS2之间存在显著相关性(P<0.001),另外TCF3表达与肿瘤组织分化程度高低、是否感染幽门螺旋杆菌有关(P<0.05)。生存分析数据表明TCF3表达量越高,胃癌患者的总生存率和进展后生存率越低(P<0.001)。生信富集分析结果显示TCF3及其相关基因在生物遗传信息保存、细胞周期、肿瘤发生发展方面有重要作用(P<0.05),蛋白质网络提示TCF3与ID1、ID2、TWIST1、MYOD1、EP300、GATA1、LYL1、TAL1、LDB1、LMO2等蛋白存在交互作用(P<0.001)。[结论]TCF3基因可作为胃癌临床预后体系中的重要生物标志物。[Objective] To investigate the expression level of TCF3 in gastric cancer tissue and its correlation with clinical prognosis.[Method]The differential expression of TCF3 in gastric cancer tissues, clinicopathological data and the relationship with mismatch repair genes were analyzed by GEPIA and ULCAN databases, and the Kaplan-Meier Plotter database was used to draw the survival and prognosis curve of TCF3 and gastric cancer patients.According to the Spearman correlation coefficient, the genes related to the role of TCF3 in gastric cancer were found.In addition, the G0/KEGG enrichment analysis of TCF3 and its related genes was performed to find the most different biological annotations.Finally, the String database was used to build the TCF3 protein and the above Regulatory interaction network of downstream proteins.[Result] The expression level of TCF3 in gastric cancer tissue was significantly higher than that in normal gastric tissue(P<0.05),and there was a significant correlation between TCF3 and mismatch repair genes MLH1,MSH2,MSH6,and PMS2(P<0.001).The expression level of TCF3 is related to the degree of differentiation and whether it is infected with Helicobacter pylori(P<0.05).Survival analysis data showed that the higher the expression of TCF3,the lower the overall survival rate and post-progression survival rate of gastric cancer patients(P<0.001).The results of bioinformatics enrichment analysis showed that TCF3 and its related genes play an important role in the preservation of biological genetic information, cell cycle, tumor occurrence and development(P<0.05).The protein network indicated that TCF3 interacted with ID1,ID2,TWIST1,MYOD1,EP300,GATA1,LYL1,TAL1,LDB1,LMO2 and other proteins(P<0.001).[Conclusion] TCF3 gene can be used as an important biomarker in the clinical prognosis system of gastric cancer.
