摘　　要：目的 探索miR-148a-3p对肺癌细胞增殖、迁移与侵袭的影响。方法 收集肺癌组织和配对癌旁组织以及肺癌细胞和正常肺上皮细胞株,RT-qPCR检测miR-148a-3p表达;Western blot和免疫组织化学检测谷氧还蛋白5(glutaredoxin 5,GLRX5)的表达。对A549细胞转染miR-148a-3p mimics和pcDNA-GLRX5后,分别用MTT、EdU、细胞划痕和Transwell实验检测细胞活性、细胞增殖、迁移和侵袭;Western blot和免疫荧光检测增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)、Ki-67、基质金属蛋白酶-2(matrix metalloproteinase-2,MMP-2)和MMP-9表达。用生物信息学和荧光素酶测定法鉴定miR-148a-3p与GLRX5的靶向关系。结果 分别与癌旁组织或正常肺上皮细胞比较,肺癌组织和肺癌细胞中miR-148a-3p表达降低,GLRX5表达升高。转染miR-148a-3p mimics后,A549细胞活性、增殖、迁移和侵袭以及PCNA、Ki67、MMP-2和MMP-9表达均降低,而过表达GLRX5能显著逆转miR-148a-3p mimics的上述作用。GLRX5是miR-148a-3p的靶基因。结论 miR-148a-3p可通过靶向GLRX5来发挥对肺癌细胞增殖、迁移和侵袭的抑制作用。Objective To explore the effects of miR-148a-3p on the proliferation,migration and invasion of lung cancer cells.Methods Lung cancer tissues and paired adjacent tissues,lung cancer cells and normal lung epithelial cell lines were collected,and the expression of miR-148a-3p was detected by RT-qPCR,the expression of glutaredoxin 5(GLRX5)was detected by Western blot and immunohistochemistry.After transfection of A549 cells with miR-148a-3p mimics and pcDNA-GLRX5,the cell viability,prolif­eration,migration and invasion were detected by MTT,EDU,cell scratch and Transwell assay,respectively;while,the expression of proliferating cell nuclear antigen(PCNA),Ki-67,matrix metalloproteinase 2(MMP-2)and MMP-9 were detected by Western blot and immunofluorescence.Bioinformatics and luciferase assay were used to identify the targeting relationship between miR-148a-3p and GLRX5.Results Compared with adjacent tissues or normal lung epithelial cells,the expression of miR-148a-3p in lung cancer tissues and lung cancer cells was decreased while the expression of GLRX5 was increased.After transfection with miR-148a-3p mimics,the viability,proliferation,migration and invasion of A549 cells,as well as the expression of PCNA,Ki67,MMP-2 and MMP-9 were de­creased,while overexpression of GLRX5 could significantly reverse the above effects of miR-148a-3p mimics.GLRX5 was the target gene of miR-148a-3p.Conclusion miR-148a-3p can inhibit the proliferation,migration and invasion of lung cancer cells by targeting GLRX5.
