摘  要:为测定独一味种子生活力及最佳染色条件,建立独一味种子活力测定的快速方法,以野生独一味种子为材料,采用2,3,5-氯化三苯基四氮唑(TTC)染色法,通过单因素试验、正交试验研究不同浸种温度、浸种时间、染色温度、染色时间和TTC浓度对独一味种子生活力测定的影响。结果表明,5个因素对独一味种子生活力的影响依次为:染色时间>染色温度>TTC浓度>浸种时间>浸种温度;正交试验染色最佳条件为25℃水浸泡9 h、0.1%TTC、40℃恒温箱染色9 h。In order to determine the viability and optimal dyeing conditions of Lamiophlomis rotata(Benth.) Kudo seeds,establish a rapid method for the determination of viability of L.rotata(Benth.) Kudo seeds,using wild L.rotata(Benth.) Kudo seeds as experimental materials,using 2,3,5-triphenyltrizole chloride(TTC) dyeing method,through univariate experiments and orthogonal experiments to study the influence of different soaking temperature,soaking time,dyeing temperature,dyeing time and TTC concentration on the determination of L.rotata(Benth.) Kudo seed vitality.The results showed that the effects of the five factors on the viability of the L.rotata(Benth.) Kudo seed from strong to small were the dyeing time,the staining temperature,the TTC concentration,the soaking time,the soaking temperature.The optimal conditions for orthogonal experimental staining were 9 hours immersion in water at 25 ℃,0.1% concentration TTC,and 40 ℃ incubator staining for 9 hours.
