摘 要:[目的]优化改善文冠果花粉离体萌发培养基。[方法]以新鲜文冠果花粉为试验材料,采用液体培养法,测定蔗糖、硼酸和钙对文冠果花粉离体萌发特性及花粉管长度的影响,筛选适合文冠果花粉离体培养液体培养基成分的最佳浓度。测定不同贮藏温度(-70、-20、4、25℃)及不同贮藏时间(2、5、10 d)下,文冠果花粉萌发率的变化。[结果]通过控制单一因素变量试验,文冠果花粉离体萌发的液体培养基最佳组分为15%蔗糖+0.02%硼酸+0.01%钙离子(硝酸钙),萌发率达92.5%,花粉管长达1.61 mm;而在花粉管生长过程中需要的钙离子浓度为0.04%。-70℃超低温贮藏花粉有利于保持花粉生活力,-20℃冷冻贮藏次之,4℃低温贮藏不利于保持花粉生活力,25℃恒温贮藏会大大降低花粉生活力。[结论]该研究可为文冠果花粉离体培养的液体培养基最佳组分配方提供参考依据,也为文冠果花粉贮藏研究奠定基础。[Objective]In order to optimize and improve the culture medium of Xanthoceras sorbifolium pollen germination in vitro.[Method]The effects of sucrose,boric acid and calcium ion on the germination rate of Xanthoceras sorbifolium pollen and pollen tube length were measured by liquid culture method,which fresh pollen was used as experimental material.So as to screen out the best concentration of liquid medium components suitable for Xanthoceras sorbifolium pollen germination in vitro.At the same time,the changes of pollen germination rate of Xanthoceras sorbifolium were measured under different storage temperatures(-70,-20,4,25℃)and different storage times(2,5,10 d).[Result]The results showed that through controlling the single factor variable test,the optimal medium group that promoted in vitro germination rate of pollen of Xanthoceras sorbifolium was 15%sucrose+0.02%boric acid+0.01%calcium ion(calcium nitrate),the germination rate was 92.5%,and the pollen tube length was 1.61 mm.While the concentration of calcium ion needed in the process of pollen tube growth was 0.04%.The statistics indicates that-70℃cryogenic storage of pollen was beneficial to keep the vitality of pollen,and 20℃refrigerated storage times,4℃low temperature storage was not conducive to maintain vitality of pollen,25℃constant temperature storage will greatly reduce the vitality of pollen.[Conclusion]This study can provide reference basis for the optimal formula of liquid medium for Xanthoceras sorbifolium pollen germination in vitro,and lay a foundation for the study of Xanthoceras sorbifolium pollen storage.
