摘  要:目的基于Wnt/β-catenin信号通路研究葛根芩连汤对结肠癌模型大鼠肠道菌群的作用机制。方法选取SD健康雄性大鼠,随机分为正常组、结肠癌组、吉西他滨组及葛根芩连汤(GQD)低、中、高剂量组,对各组大鼠进行腹泻、便血、大便性状等肠炎症状评分,采用鲎试剂显色基质法测定血清内毒素脂多糖(LPS)水平,采用酶联免疫吸附实验法检测肿瘤坏死因子-α(TNF-α)、白细胞介素-10(IL-10)、白细胞介素-6(IL-6)炎性因子水平,通过肠道内容物中微生物的培养检测大肠埃希菌、肠球菌、乳酸杆菌及双歧杆菌数量,采用Shannons法检测肠道内容物菌群的多样性,采用Western blot法检测Wnt/β-catenin通路蛋白相对表达量。结果与正常组相比,结肠癌组、吉西他滨组及GQD低、中、高剂量组大鼠TNF-α、IL-10、IL-6、LPS水平及腹泻、便血、大便性状症状评分升高,差异具有统计学意义(P<0.05);与结肠癌组相比,GQD低、中、高剂量组大鼠TNF-α、IL-10、IL-6、LPS水平及腹泻、便血、大便性状症状评分依次降低,差异具有统计学意义(P<0.05)。与正常组相比,结肠癌组、吉西他滨组及GQD低、中、高剂量组大鼠大肠埃希菌、肠球菌数量,肠道菌群多样性分析中S值、E值、H值以及Wnt/β-catenin通路蛋白水平升高,乳酸杆菌及双歧杆菌数量降低,差异具有统计学意义(P<0.05);与结肠癌组相比,GQD低、中、高剂量组大鼠肠道内容物菌群的S值、E值、H值均降低,差异具有统计学意义(P<0.05)。结论葛根芩连汤能有效改善腹泻、便血、大便性状等症状,减轻LPS及炎症反应,降低大肠埃希菌、肠球菌、乳酸杆菌、双歧杆菌数量及肠道菌群数值,并升高Wnt/β-catenin通路蛋白水平,Wnt/β-catenin通路蛋白可有效减轻肠道反应,起到肠道保护作用。Objective To server the mechanism of Gegen Qinlian Decoction(葛根芩连汤,GQD)on intestinal flora in colon cancer model rats based on Wnt/β-catenin signaling pathway.Methods Selected from SD healthy male rats,randomized into normal group,colon cancer group,gemcitabine group and low,medium and high dose of GQD groups,all groups of rats were scored for diarrhea,stool stool,stool traits and other enteritis symptoms,the serum levels of endotoxin lipopolysaccharide(LPS)were determined by Lmatrix,the levels of tumor necrosis factor-α(TNF-α),interleukin-10(IL-10),and interleukin-6(IL-6)were measured by enzyme-linked immunosorbent assay,the numbers of Escherichia.coli,Enterococcus,Lactobacillus and Bifidobacterium were detected by the culture of the microorganisms in the intestinal contents,the diversity of the intestinal contents was detected by Shannons,relative expression of Wnt/β-catenin pathway proteins was determined by Western blot assay.Results Compared with the normal group,the levels of TNF-α,IL-10,IL-6,LPS,and the scores of diarrhea,blood dose the stool,and stool traits were increased dose the colon cancer group,gemcitabine group,and GQD low-,medium-,and high dose groups.The difference was statistically significant(P<0.05);compared with the colon cancer group,the levels of TNF-α,IL-10,IL-6,LPS and the symptom scores of diarrhea,blood in the stool and stool of the rats in the GQD low,medium and high dose groups decreased sequentially,and the difference was statistically significant(P<0.05).Compared with the normal group,the numbers of Escherichia coli and Enterococcus in colon cancer group,gemcitabine group and GQD low,medium and high dose groups,S value,E value,H value in the analysis of intestinal flora diversity and Wnt/β-catenin pathway protein level increased,and the number of lactobacillus and Bifidobacteriun decreased,the difference was statistically significant(P<0.05).The S value,E value and H value of intestinal flora were all decreased,and the difference was statistically significant(P<0.05
