摘 要:目的评价以酸枣仁、天麻为主的复方口服液改善睡眠的作用及机制。方法以酸枣仁、天麻为主的复方口服液按照低(19.5 mL/kg)、中(39 mL/kg)、高(117 mL/kg)3个剂量每天给予小鼠灌胃,阴性对照物为生理盐水,连续给药30 d后通过直接睡眠实验、延长戊巴比妥钠睡眠时间实验、戊巴比妥钠阈下剂量催眠实验和巴比妥钠睡眠潜伏期实验来观察小鼠睡眠相关指标,并采用ELISA法测定脑组织内GABA、5-HT、DA的水平。结果与阴性对照组相比,复方口服液3个剂量组对小鼠体质量无影响,均无直接睡眠作用,可显著延长小鼠在戊巴比妥钠睡眠实验中的睡眠时间(P<0.001或0.01),均能增加戊巴比妥钠阈下剂量诱导的小鼠睡眠发生率,均能显著缩短小鼠巴比妥钠睡眠潜伏期(P<0.001)。不同剂量组均能提高小鼠脑组织内神经递质GABA(P<0.001或0.01)、5-HT(P<0.001)的水平;低、中剂量虽能提高多巴胺的含量,但差异没有统计学意义,高剂量能显著提高和多巴胺(P<0.001)的水平。结论在实验条件和所设剂量下,实验结果显示该复方口服液对睡眠具有明显的改善作用,其作用机制可能与提高脑组织中GABA、5-HT、DA的水平有关。Objective To evaluate the sleep-improving effect and mechanism of compound oral liquid based on Suanzaoren(Ziziphi Spinosae Semen)and Tianma(Gastrodiae Rhizoma).Methods The compound oral liquid based on Suanzaoren(Ziziphi Spinosae Semen)and Tianma(Gastrodiae Rhizoma)was administered orally to mice in three doses of low(19.5 mL/kg),medium(39 mL/kg),and high(117 mL/kg)every day.The negative control was physiological Saline,after 30 days of continuous administration,observe the sleep-related indicators of mice through direct sleep test,pentobarbital sodium sleep prolongation test,pentobarbital sodium subthreshold dose hypnosis test and barbital sodium sleep latency test.And the levels of GABA,5-HT,and DA in brain tissues were determined by ELISA.Results Compared with the negative control group,three doses of oral liquid for sleep improvement had no effect on the body weight of mice,none of them had a direct sleep effect;it can significantly prolong the sleep time of mice in the pentobarbital sodium sleep experiment(P<0.001 or 0.01);both can increase the incidence of sleep in mice induced by the subthreshold dose of pentobarbital sodium;both can significantly shorten the sleep latency of mouse barbital sodium(P<0.001).Different dose groups can increase the levels of neurotransmittersγ-aminobutyric acid(P<0.001 or 0.01)and 5-hydroxytryptamine(P<0.001)in mouse brain tissue.Low and medium doses could increase the level of dopamine,but the difference was not statistically significant.High doses could significantly increase the level of dopamine and dopamine(P<0.001).Conclusion Under the conditions of this experiment and the dose set,the results of this experiment showed that the sleep-improving oral liquid has a significant improvement effect on sleep,and its mechanism of sleep improvement may be related to increasing the levels of GABA,5-HT,and DA in brain tissue.
