摘  要:目的:采用PIWI-RNA(piRNA)表达谱技术筛选原发性高血压病患者与正常健康人群血浆中差异性表达的piRNAs;运用实时荧光定量PCR(qRT-PCR)技术对表达谱中差异表达的piRNAs进行验证;选取一个高表达piRNA并分析与高血压病患者临床资料的关联性,为探索piRNA作为高血压发病的重要环节奠定理论基础。方法:收集5例高血压病患者和5例健康人血浆中的总RNA,运用piRNA表达谱技术检测两组中piRNAs的表达情况,设定筛选条件筛选出差异表达的piRNAs。在72例高血压患者及72例对照者血浆样本中应用qRT-PCR技术对5个表达上调和5个表达下调的piRNAs进行验证,并明确其表达情况。收集高血压病患者的临床资料,分析piR-37390与临床病例资料的关联性。结果:piRNA表达谱结果显示,与健康人群组相比,高血压病患者血浆中差异表达的piRNAs有4845种,其中2322种piRNAs表达上调,2523种表达下调。当设定P值<0.01且差异倍数>3倍条件后选出5个高表达的piRNAs和5个低表达的piRNAs,再应用qRT-PCR实验验证1种表达上调的piRNA(piR-37390)以及2种表达下调的piRNAs(piR-43926和piR-45076),P值均<0.05。统计学分析piR-37390与高血压病患者临床资料的关系,得出piR-37390表达与高血压病级别(P=0.000)、HDL-C(P=0.001)、肾素(P=0.032)和醛固酮(P=0.028)密切相关(均P<0.05),与血管紧张素Ⅱ几乎有关联(P=0.053),但是与患者年龄、性别、BMI、TG、LDL-C、COR和ACTH均无关联(均P>0.05)。结论:本实验收获了高血压病患者血浆中piRNAs表达谱特征,并验证了高血压病患者血浆中piR-37390表达上调;piR-37390与高血压病的发生可能有关联,参与或促进了高血压病的形成;为piRNAs作为高血压病的形成和治疗奠定理论基础,提供了新的思路。Objective:To screen the differentially expressed piRNAs in plasma of patients with essential hypertension and normal healthy people by using PIWI-RNA(piRNA)expression profile technology.Real time fluorescent quantitative PCR(qRT-PCR)was used to verify the differentially expressed piRNAs in the expression profile.A high expression piRNA was selected and analyzed the correlation with the clinical data of patients with hypertension,so as to lay a theoretical foundation for exploring piRNA as an important link in the pathogenesis of hypertension.Methods:The total RNA in plasma of 5 patients with hypertension and 5 healthy people was collected,and the expression of piRNAs in the two groups was detected by piRNA expression profile technology.The screening conditions were set to screen out the differentially expressed piRNAs.In the plasma samples of 72 hypertensive patients and 72 controls,the expression of 5 up-regulated and 5 down-regulated piRNAs was verified by qRT-PCR,and their expression was determined.The clinical data of patients with hypertension were collected and the correlation between piR-37390 and clinical case data was analyzed.Results:The results of piRNA expression profile showed that there were 4845 kinds of piRNAs differentially expressed in the plasma of patients with hypertension compared with the healthy group,of which 2322 kinds of piRNAs were up-regulated and 2523 kinds were down regulated.When the conditions of P value<0.01 and difference multiple>3 times were set,5 highly expressed piRNAs and 5 low expressed piRNAs were selected,and then qRT-PCR was used to verify that 1 up-regulated piRNA(piR-37390)and 2 down-regulated piRNAs(piR-43926 and piR-45076)had P values<0.05.Statistical analysis of the relationship between piR-37390 and the clinical data of patients with hypertension showed that the expression of piR-37390 was closely related to the grade of hypertension(P=0.000),HDL-C(P=0.001),renin(P=0.032)and aldosterone(P=0.028)(all P<0.05),almost related to Aldosterone(P=0.053),but not related to
