摘  要:薰衣草是新疆当地主要的特色经济作物,为了扩大薰衣草精油的产品加工范围,提升其开发利用价值,应用气相色谱-质谱联用仪对EO1和EO22种精油的主成分进行了检测,并选取BALB/c小鼠为研究对象,通过空瓶刺激+咖啡因注射营造小鼠失眠模型,以嗅闻精油的方式进行干预,选择DZP作为阳性对照,采用高架十字迷宫、戊巴比妥诱导睡眠评估作用效果,采集小鼠血液和脑组织,检测其中Cortisol、GABA、ACH和HIS等的含量,探究以薰衣草为主的精油EO1和EO2针对焦虑和咖啡因协同引起失眠的缓解效果及作用机理。结果表明:精油EO1共鉴定出65种精油成分,达到检出成分的97.48%,EO2共鉴定出74种精油成分,达到检出成分的97.42%,2种精油的主要成分均为乙酸芳樟酯、芳樟醇、柠檬稀、乙酸薰衣草酯;2个精油组小鼠进入开放臂的频率均为33.33%,且EO2组进入开放臂的次数和停留时间以及活动的总路程均高于模型组,分别为14.58%、4.33%和(15855.98±949.89)mm,但差异不显著;戊巴比妥诱导睡眠试验中,2个精油组小鼠的入睡率均高于模型组在80%以上,2种精油对入睡潜伏期、睡眠持续时间和睡眠片段的影响有差异,EO1分别为(445±86.85)s、(1284.40±431.10)s和1,而EO2为(1177±107.08)s、(665.25±180.30)s和3,从睡眠行为上分析EO1作用效果稍优于EO2;神经递质/神经调节剂的检测中,2个精油组对这些成分含量的改变均大于阳性药物组,其中对GABA、DA、NE和HIS的改变达到了显著性差异,P<0.05或P<0.01。由此可见,2种精油对焦虑与失眠都有一定的缓解效果,而且针对焦虑协同咖啡因致小鼠失眠的缓解可能是通过对Cortisol、GABA和ACH等神经递质/神经调节剂的调节发挥作用。该研究结果为后续进一步揭示薰衣草精油作用机理、拓宽精油应用领域提供了参考。Lavender is the main characteristic economic crop in Xinjiang.In order to expand the product processing range of lavender essential oil and improve its development and utilization value,the main components of EO1 and EO2 essential oils were detected by gas chromatography-mass spectrometry.BALB/c mice were selected as the research object.The mouse insomnia model was created by empty bottle stimulation&caffeine injection.The intervention was conducted by smelling the essential oil.DZP was selected as the positive control.The elevated cross maze and pentobarbital were used to inducing sleep in order to evaluate the effect.The blood and brain tissues of mice were collected,and the contents of Cortisol,GABA,ACH and HIS were detected.The experiment was performed to explore the effect and mechanism of the essential oils EO1 and EO2,mainly lavender,on anxiety and caffeine induced insomnia.The results showed that 65 essential oil components were identified by EO1,accounting for 97.48%of the detected components.74 essential oil components were identified by EO2,accounting for 97.42%of the detected components.The main components of the two essential oils were linalool acetate,linalool,limonin,and lavender acetate.The frequencies of mice entering the open arm in the two essential oil groups were both 33.33%while the number of times,residence time and total distance of activity in the EO2 group were higher than those in the model group,14.58%,4.33%and(15855.98±949.89)mm,respectively,but the difference was not significant.In the pentobarbital induced sleep experiment,the sleep rate of the two essential oil groups was higher than that of the model group by more than 80%.The effects of the two essential oils on sleep latency,sleep duration and sleep fragments were different,with EO1 being(445±86.85)s,(1284.40±431.10)s and 1 respectively,and EO2 being(1177±107.08)s,(665.25±180.30)s and 3 respectively.From the perspective of sleep behavior,the effect of EO1 was slightly better than EO2.In the detection of neurotransmitter/neu
