摘  要:【目的】监测河南省猪细小病毒1型(porcine parvovirus 1,PPV1)的变异情况。【方法】采集自河南省鹤壁市和安阳市初产母猪的流产胎儿阳性样品,利用PCR方法鉴定获得2株PPV1流行毒株,经过全基因组测序,分别命名为CH/HN-H/2021和CH/HN-3/2021。通过分析同源性和构建遗传进化树分析PPV1全基因组、NS1基因、VP2基因的变异情况,并且比对分析NS1蛋白和VP2蛋白的氨基酸变异情况及非编码区基因缺失情况。【结果】2条序列的核苷酸相似性为99.9%;与46株国内外PPV1流行毒株全基因组的核苷酸同源性为93.5%~99.9%;NS1基因核苷酸同源性为98.6%~99.9%,NS1蛋白氨基酸同源性为98.2%~99.9%;VP2基因核苷酸同源性为98.4%~99.9%,VP2蛋白氨基酸同源性为98.3%~99.9%,说明本研究鉴定的毒株呈现遗传进化稳定性。本研究鉴定株与湖南及吉林毒株亲缘关系最近,而与河南原有毒株亲缘关系较远。在对NS1蛋白与VP2蛋白的氨基酸变异分析中发现,NS1蛋白发生6处突变,VP2蛋白发生7处突变,均在经典突变位点范围内,符合PPV1遗传变异的保守性。鉴定株VP2基因后部非编码区基因与NADL-2株相比缺失127 nt。【结论】本研究鉴定的毒株呈现遗传进化稳定性,河南省存在新旧PPV1毒株同时流行的情况。【Objective】The study was conducted to monitor the variation of porcine parvovirus 1 in Henan Province.【Method】The positive samples of aborted fetuses from newborn sows in Hebi City and Anyang City,Henan Province were collected.Two PPV1 epidemic strains were identified by PCR method.After complete genome sequencing,the resulting two strains were named CH/HN-H/2021 and CH/HN-3/2021 respectively.Through homology and genetic evolution tree construction,the genetic evolution of PPV1 complete genome,NS1 gene and VP2 gene was analyzed,and the amino acid variation of NS1 protein and VP2 protein was compared and analyzed.【Result】The nucleotide homology of the two sequences was 99.9%,while the complete genome nucleotide homology with 46 domestic and international strains was 93.5%-99.9%.The NS1 gene nucleotide homology was 98.6%-99.9%,the amino acid homology was 98.2%-99.9%,the homology of VP2 gene was 98.4%-99.9%,and the amino acid homology was 98.3%-99.9% indicating that the strains identified in this study showed genetic and evolutionary stability.The strains identified in this study have the closest genetic relationship with Hunan and Jilin strains,but have a far genetic relationship with the original strains in Henan.There is a situation that the strains identified in this study and the original strains in Henan are prevalent at the same time.In the analysis of amino acid variation of NS1 protein and VP2 protein,it was found that the mutation point of NS1 protein has 6 mutations and VP2 protein has 7 mutations,all within the range of classical mutation sites,in accordance with the conservatism of PPV1 genetic variation.The gene in the posterior non-coding region of the identified strain VP2 gene was missing 127 nt compared to NADL-2.【Conclusion】The strains identified in this study showed genetic and evolutionary stability,and there were new and old PPV1 strains prevalent at the same time in Henan Province.
