摘　　要：第二信使环磷酸腺苷(cyclic adenosine monophosphate,cAMP)参与多种神经退行性疾病的发生发展,如阿尔兹海默病、帕金森病以及亨廷顿病。但是对cAMP进行直接调控具有一定的难度,因此靶向调控cAMP的水解酶有望成为改善神经退行性疾病的新策略。磷酸二酯酶4(phosphodiesterases, PDE4)是特异性催化c AMP的酶,已被证实在多种神经退行性疾病的发展中具有重要作用。研究并开发靶向PDE4的药物将有助于神经退行性疾病的治疗。本文综述了PDE4的特性和功能,回顾了其在神经退行性疾病中的作用及研究进展,并对其在神经退行性疾病中的潜在治疗作用作出展望。Aberrant changes in the second messenger cyclic adenosine monophosphate(cAMP)are involved in the development of several neurodegenerative diseases such as Alzheimer's disease,Parkinson's disease and Huntington's disease.However,it is difficult to directly regulate cAMP signal.Therefore,targeted regulation of cAMP hydrolase is expected to be a new strategy to improve neurodegenerative diseases.Phosphodiesterase 4(PDE4),an enzyme that specifically catalyzes cAMP,has been shown to have an important role in the development of a variety of neurodegenerative diseases.Research into drugs targeting PDE4 will help in the treatment of neurodegenerative diseases.In this paper,we will describe the characteristics and function of PDE4,review its role and research into progress in neurodegenerative diseases and future development trend of its potential therapeutic effect in neurodegenerative diseases.
