摘 要:目的基于表皮生长因子受体(EGFR)/血管内皮生长因子A(VEGF-A)信号通路探讨川芎挥发油对胶质瘤血管生成的抑制作用。方法实验分为对照组、20μmol/L川芎挥发油组、40μmol/L川芎挥发油组、80μmol/L川芎挥发油组。各组胶质瘤U87细胞培养24 h后,镜下观察细胞形成血管生成拟态情况,CCK-8法检测细胞存活率,Transwell测定细胞迁移和侵袭能力,PCR技术和Western blot技术检测EGFR、VEGF-A mRNA和蛋白表达情况。结果川芎挥发油各组血管拟态计数均明显少于对照组(P均<0.05),细胞存活率均明显低于对照组(P均<0.05),穿过Transwell小室的细胞数和穿过含基质胶Transwell小室的细胞数均明显少于对照组(P均<0.05),细胞中EGFR、VEGF-A mRNA和蛋白相对表达量均明显低于对照组(P均<0.05),且川芎挥发油各组上述各指标(除VEGF-A蛋白相对表达量)均呈剂量依赖性减少或降低。结论川芎挥发油可呈浓度依赖性抑制胶质瘤血管生成和细胞迁移、侵袭,机制可能与抑制EGFR/VEGF-A信号通路有关。Objective It is to explore the inhibitory effect of volatile oil from Rhizoma Ligustici Chuanxiong on glioma angiogenesis based on epidermal growth factor receptor(EGFR)/vascular endothelial growth factor A(VEGF-A)signaling pathway.Methods The experiment was divided into control group,20μmol/L Rhizoma Ligustici Chuanxiong volatile oil group,40μmol/L Rhizoma Ligustici Chuanxiong volatile oil group,and 80μmol/L Rhizoma Ligustici Chuanxiong volatile oil group.After the U87 glioma cells in each group were treated for 24 h,the ability of glioma cells to form vasculogenic mimicry was observed under microscope,the cell survival was detected by CCK-8 method,the cell migration and invasion ability were detected by Transwell,the EGFR,VEGF-A mRNA and protein expressions were detected by PCR technique and Western blot technique,respectively.Results Compared with the control group,the vascular mimicry counts were significantly less,the cell survival rates were significantly lower,the number of cells passing through the Transwell and the number of cells passing through the matrix gel-containing Transwell were significantly less,and the relative expression of EGFR,VEGF-A mRNA and protein in the cells were significantly lower in each group of Rhizoma Ligustici Chuanxiong volatile oil,the differences were all significant(all P<0.05),and all of the above indicators(except the relative expression of VEGF-A protein)were reduced or decreased in a dose-dependent manner.Conclusion Rhizoma Ligustici Chuanxiong volatile oil can inhibit glioma angiogenesis,cell migration and invasion in a concentration-dependent manner,and the mechanism may be related to the inhibition of EGFR/VEGF-A signaling pathway.
