摘  要:目的 探讨枫蓼肠胃康颗粒对肥胖诱导的2型糖尿病大鼠糖脂代谢及胰高血糖素样肽-1(GLP-1)的影响。方法 7周龄SD大鼠70只,随机取10只作为分为空白组,给予正常饲料喂养;其余大鼠采用高脂高糖饲料喂养结合腹腔注射链脲佐菌素方法建立2型糖尿病模型。将造模成功2型糖尿病大鼠随机分为模型组、西格列汀组及枫蓼肠胃康低、中、高剂量组,每组10只。西格列汀组给予西格列汀16.7 mg/kg灌胃,枫蓼肠胃康低、中、高剂量组分别给予2.65 g/kg、5.3 g/kg、10.6 g/kg枫蓼肠胃康颗粒灌胃,空白组和模型组给予生理盐水灌胃,均1次/d,连续4周。测量各组大鼠造模结束和灌胃4周后体重,检测各组大鼠灌胃4周后血糖、血清胰岛素、血脂[胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)]及血清GLP-1水平,免疫组织化学法检测回肠组织中GLP-1表达情况。结果 灌胃4周后,枫蓼肠胃康中、高剂量组及西格列汀组大鼠体重与造模后比较差异无统计学意义(P均>0.05);模型组与枫蓼肠胃康低剂量组大鼠体重与造模后比较均明显降低(P均<0.05)。枫蓼肠胃康中、高剂量组和西格列汀组大鼠空腹血糖、负荷后30 min血糖、负荷后60 min血糖、负荷后120 min血糖和空腹胰岛素水平均明显低于模型组(P均<0.05);枫蓼肠胃康高剂量组和西格列汀组大鼠负荷后60 min和120 min血糖均明显低于枫蓼肠胃康低剂量组(P均<0.05);枫蓼肠胃康中剂量组和西格列汀组大鼠负荷后60 min胰岛素水平均明显高于枫蓼肠胃康低剂量组(P均<0.05)。枫蓼肠胃康高剂量组和西格列汀组大鼠血清TC、TG、LDL-C水平及枫蓼肠胃康中剂量组大鼠血清TC、TG水平均明显低于模型组(P均<0.05),枫蓼肠胃康中剂量组大鼠HDL-C水平明显高于模型组(P均<0.05)。枫蓼肠胃康中、高剂量组和西格列汀组血清GLP-1水平均明显高于模型组(P�Objective It is to investigate the effect of Feng-Liao-Chang-Wei-Kang(FLCWK)granules on glycolipid metabolism and glucagon like peptide-1(GLP-1)in obesity induced type 2 diabetic rats.Methods Seventy 7-week-old SD rats were selected,in which 10 rats were randomly selected as a blank group and fed with normal diet,the rest rats were fed with high-fat and high-sugar diet combined with intraperitoneal injection of streptozotocin to establish type 2 diabetic models.The successfully modeled type 2 diabetic rats were randomly divided into model group,sitagliptin group and FLCWK granules low,medium and high dose groups,with 10 rats in each group.The sitagliptin group was given sitagliptin 16.7 mg/kg by gavage,the FLCWK granules low,medium and high dose groups were given FLCWK granules 2.65 g/kg,5.3 g/kg and 10.6 g/kg by gavage,respectively,and the blank group and model group were given physiological saline by gavage,all once a day,continuously treated for 4 weeks.The body weight of the rats in each group was measured at the end of modeling and after 4 weeks of intragastric administration,and the levels of blood glucose,serum insulin,blood lipids[cholesterol(TC),triacylglycerol(TG),low-density lipoprotein cholesterol(LDL-C),high-density lipoprotein cholesterol(HDL-C)]and serum GLP-1 in the rats were measured after 4 weeks of intragastric administration,the expression of GLP-1 in ileum tissue was detected by immunohistochemistry.Results After 4 weeks of gavage,no statistically significant changes of body weight were found in the FLCWK granules medium and high dose groups and sitagliptin group compared with those after modeling(all P>0.05),the body weights of the rats in the model group and the FLCWK granules low-dose group were significantly reduced compared with those after modeling(all P<0.05).The levels of fasting blood glucose,30 min post-load blood glucose,60 min post-load blood glucose,120 min post-load blood glucose and fasting insulin of the rats in the FLCWK granules medium and high dose groups and the sitaglipti
