摘 要:目的:利用癌症基因组图谱的头颈鳞癌数据探讨可变剪接与肿瘤微环境的关系。方法:从UCSC-Xena、TIMER2.0、Spliceseq网站分别下载TCGA头颈鳞癌的转录组和临床数据、免疫浸润数据以及剪接百分比数据。根据剪接数据对肿瘤样本进行分型,对各型进行通路富集及肿瘤微环境分析。结果:共纳入485例肿瘤和41例正常样本,涉及剪接事件6 720个,基因3 330个。一致性聚类将头颈鳞癌分成4个亚型。亚型间的预后及部分临床特征差异显著。4个亚型间有不同的微环境特征和检查点基因特征。结论:头颈鳞癌的可变剪接与临床表型及肿瘤微环境密切相关。基于可变剪接的头颈鳞癌分型有一定的免疫靶点选择和研究的提示作用。Objective:To explore the relationship between alternative splicing and tumor microenvironment by using the data of head and neck squamous cell carcinoma(HNSCC)in the cancer genome map.Methods:The transcription and clinical data,immune infiltration data and splicing percentage data of TCGA HNSCC were downloaded from UCSC-Xena,TIMER2.0 and Spliceseq websites.Tumor samples were classified according to splicing data,and pathway enrichment and tumor microenvironment analysis were performed for each tumor type.Results:A total of 485 tumors and 41 normal samples were included,involving 6720 splicing events and 3330 genes.Consistency clustering divided HNSCC into 4 subtypes.There was significant difference in prognosis and some clinical features among the subtypes.The 4 subtypes had different microenvironment characteristics and checkpoint gene characteristics.Conclusion:The alternative splicing changes of HNSCC were closely related to clinical phenotype and microenvironment.The subtyping of HNSCC based on alternative splicing had certain implications for immune target selection and research.
