摘 要:目的探讨TGF-β信号通路关键分子转化生长因子β-1型受体(transforming growth factor-βreceptor 1,TβR1)和磷酸化Smad2/3(phosphorylated Smad2/3,p-Smad2/3)蛋白在哈萨克族(哈族)食管鳞状细胞癌(鳞癌)中的表达及其临床意义。方法采用GEO和TCGA数据库分析食管鳞癌标本与癌旁正常组织中TβR1和p-Smad2/3的表达;运用免疫组织化学技术检测127例哈族食管鳞癌组织和82例癌旁正常组织中TβR1和p-Smad2/3的蛋白表达状况,分析及探讨TβR1、p-Smad2/3蛋白表达水平与哈族食管鳞癌患者临床病理参数及预后的关系。结果TβR1和p-Smad2/3蛋白在哈族食管鳞癌中表达上调(P=4.79×10^(-7);P=2.21×10^(-9)),且两者的表达呈正相关(r=0.322,P=0.002);TβR1、p-Smad2/3蛋白表达水平与食管鳞癌的浸润深度密切相关(P=0.003;P=0.005),且有淋巴结转移的样本中TβR1、p-Smad2/3蛋白表达水平明显高于无淋巴结转移的样本(P=0.028;P<0.001),而TβR1、p-Smad2/3蛋白表达与食管鳞癌患者年龄、性别和肿瘤分化程度无明显相关(均P>0.05);Kaplan-Meier生存分析显示TβR1和p-Smad2/3蛋白高表达食管鳞癌患者生存时间明显短于低表达患者(P=0.0013;P=0.0017)。结论TβR1和p-Smad2/3蛋白表达与哈族食管鳞癌患者的肿瘤浸润深度、淋巴结转移及预后密切相关。Objective To investigate the expression and clinical significance of transforming growth factorβ-1 receptor(TβR1)and phosphorylated Smad2/3 proteins(p Smad2/3)which are the key factors of TGF-βsignaling pathway,in esophageal squamous cell carcinoma(ESCC)among Kazakh population.Methods The expression levels of TβR1 and p Smad2/3 in ESCC specimens and adjacent normal tissues were analyzed by GEO and TCGA database.Immunohistochemistry was used to detect the protein expression of TβR1 and p Smad2/3 in 127 Kazakh ESCC tissues and 82 adjacent normal tissues.The relationship between the protein expression levels of TβR1 and p Smad2/3,clinicopathological parameters and prognosis of Kazakh ESCC patients was analyzed and explored.Results Expression levels of TβR1 and p Smad2/3 protein were upregulated(P=4.79×10^(-7);P=2.21×10^(-9))and positively correlated(r=0.322,P=0.002)in Kazakh ESCC.The expression levels of TβR1 and p Smad2/3 protein were closely correlated with the invasion depth(P=0.003;P=0.005)and lymph node metastasis(P=0.028;P=3×10-6)of ESCC,and there was no significant difference between TβR1 and p Smad2/3 protein expression and age,sex and tumor differentiation of ESCC patients(P>0.05);Kaplan-Meier analysis showed that the survival time of patients with high ex-pression of TβR1 and p-Smad2/3 proteins was significantly shorter than that of patients with low expression(P=0.0013;P=0.0017).Conclusion The expression levels of TβR1 and p Smad2/3 protein are closely correlated with the depth of invasion,lymph node metastasis and prognosis of Kazakh ESCC.
