FGF21过表达抑制胰腺癌细胞增殖、迁移和侵袭并促进细胞凋亡

摘　　要：目的 探讨成纤维细胞生长因子21(FGF21)基因过表达对人胰腺癌细胞株PANC-1凋亡、增殖、迁移和侵袭的影响。方法 将含FGF21基因序列的重组慢病毒载体FGF21-3xflag-ZsGreen-PURO和空载体HBLV-ZsGreen-PURO感染人胰腺癌PANC-1细胞,采用Western blot和RT-qPCR检测过表达FGF21的PANC-1细胞中FGF21、Caspase3、Cleaved-caspase3、Caspase9蛋白和FGF21、肿癌坏死因子-α(TNF-α)、白细胞介素-6(IL-6)的mRNA表达,流式细胞仪检测过表达FGF21的PANC-1细胞的凋亡,CCK-8法检测其增殖能力,划痕愈合实验和Transwell实验检测其迁移和侵袭能力。结果 HBLV-h-FGF21-3xflag-ZsGreen-PURO转染人胰腺癌细胞株PANC-1后,FGF21蛋白表达水平较空载体感染组升高,凋亡蛋白Caspase3、Cleavedcaspase3、Caspase9蛋白的表达水平均升高(P均<0.01);FGF21的mRNA表达水平较空载体感染组升高,且TNF-α和IL-6的mRNA表达水平降低(P均<0.01);FGF21过表达后可增加吉西他滨诱导的胰腺癌细胞株PANC-1凋亡率,增殖、迁移和侵袭能力均降低(P均<0.01)。结论 FGF21可抑制胰腺癌细胞增殖、迁移和侵袭并促进细胞凋亡。Objective To investigate the effects of overexpression of fibroblast growth factor 21(FGF21)gene on apoptosis,proliferation,migration and invasion of human pancreatic cancer cell line PANC-1.Methods Recombinant lentiviral vector HBLV-h-FGF21-3xflag-ZsGreen-PURO and empty vector HBLV-ZsGreen-PURO containing FGF21 gene were transfected into human pancreatic cancer PANC-1 cells.The mRNA expressions of FGF21,Caspase3,Cleaved-caspase3,Caspase9 and FGF21,TNF-a and IL-6 in PANC-1 cells overexpressing FGF21 were detected by Western blot and RT-qPCR.Apoptosis of PANC-1 cells overexpressing FGF21 was detected by flow cytometry,proliferation was detected by CCK-8 assay,migration and invasion ability were detected by scratch healing assay and Transwell assay.Results After transfection of human pancreatic cancer cell line PANC-1 with HBLV-h-FGF21-3xflag-ZsGreen-PURO,the expression level of FGF21 protein was higher than that of the empty vector infected group,and the expression levels of apoptosis protein Caspase3,Cleaved-caspase3,and Caspase9 were also higher(P all<0.01).The mRNA expression level of FGF21 was higher than that of the empty vector infected group,and the mRNA expression levels of TNF-a and IL-6 were lower(P all<0.01).FGF21 overexpression increased the apoptosis rate of gemcitabine induced pancreatic cancer cell line(PANC-1),and decreased the proliferation,migration and invasion ability(P all<0.01).Conclusion Fibroblast growth factor 21 can inhibit the proliferation,migration and invasion of pancreatic cancer cells and promote cell apoptosis.
