摘 要:目的探讨二氮嗪能否在大鼠肠道缺血再灌注模型中发挥保护作用。方法将24只8周龄健康雌性SD大鼠按随机数字表法分为3组(每组8只):空白组、模型组和二氮嗪组。二氮嗪组在小肠缺血第30 min时腹腔注射二氮嗪(10 mL/kg);模型组在相同时间点腹腔注射9 g/L盐水(10 mL/kg)。小肠缺血持续30 min,再灌注2 h后取材。采用酶联免疫吸附试验(ELISA)检测血清中丙二醛(MDA)水平;采用Western blot蛋白质印迹法检测肠黏膜紧密连接蛋白Claudin-1、Occludin含量以及肠黏膜内质网应激蛋白葡萄糖调节蛋白78(GRP-78)、半胱天冬蛋白酶12(Caspase-12)含量。结果模型组血清MDA水平高于空白组及二氮嗪组,空白组与二氮嗪组血清MDA水平未见明显差异。二氮嗪组肠黏膜Claudin-1水平高于空白组及模型组,且差异有统计学意义。3组肠黏膜Occludin及GRP-78水平未见明显差异。二氮嗪组及模型组肠黏膜上皮细胞Caspase-12水平低于空白组,两组比较差异有统计学意义。结论二氮嗪能通过降低血清MDA水平,提高大鼠肠黏膜上皮细胞紧密连接蛋白Claudin-1水平从而减轻缺血再灌注对肠道的损伤。Objective To investigate whether diazoxide can play a protective role in rat intestinal ischemia-reperfusion model.Methods Twenty-four 8-week-old healthy female SD rats were randomly divided into 3 groups(8 rats in each group):blank group,model group and diazoxide group.In the diazoxide group,diazoxide(10 ml/kg)were injected intraperitoneally at 30 min of intestinal ischemia.The model group was intraperitoneally injected with 9 g/L saline(10 ml/kg)at the same time point.The intestinal ischemia lasted for 30 min,and the samples were taken after reperfusion for 2 hours.The level of malondialdehyde(MDA)in serum was detected by enzyme-linked immunosorbent assay(ELISA).The contents of tight junction proteins,Claudin-1 and Occudin in intestinal mucosa and the contents of endoplasmic reticulum stress protein glucose-regulated protein 78(GRP-78)and Caspase-12 in intestinal mucosa were detected by Western blot.Results The serum MDA level in the model group was higher than that in the blank group and the diazoxide group.There was no significant difference in serum MDA level between the blank group and the diazoxide group.The level of Claudin-1 in intestinal mucosa of diazoxide group was higher than that of blank group and model group,and the difference was statistically significant.There was no significant difference in the levels of Occludin and GRP-78 in the intestinal mucosa of the three groups.The level of Caspase-12 in intestinal mucosal epithelial cells of the diazoxide group and the model group was lower than that of the blank group,and the difference between the two groups was statistically significant.Conclusion Diazoxide can reduce the intestinal injury caused by ischemia-reperfusion by reducing serum MDA level and increasing the level of tight junction protein Claudin-1 in intestinal mucosal epithelial cells of rats.
