摘　　要：目的 探讨PTEN基因及P53基因在结直肠癌中的表达与分布,分析其临床意义。方法 收集59例结直肠癌和28例结直肠良性病变组织,采用免疫组织化学SABC染色法,染色观察PTEN及P53基因的表达和分布。结果 PTEN基因及P53基因在结直肠癌组织中的表达,低分化高于中高分化和良性病变组织及癌前病变组织。阳性表达平均光密度值(AOD)PTEN为(0.512±0.016),P53为(0.326±0.028);在结直肠良性病变组织中,PTEN基因及P53基因的平均光密度值则分别为(0.438±0.047)及(0.251±0.018)。差异均有统计学意义(P<0.05)。患者的年龄、性别、Ducks分期与PTEN基因及P53基因的表达水平无关,但二者基因的表达水平与恶性肿瘤的分化程度有关。结论 结直肠癌的分化程度越低,PTEN基因和P53基因表达越高。可作为结直肠癌肿瘤转移及预后监测的一项重要指标。Objective To explore the expression of phosphatase and tensin homolog deleted on PTEN gene and P53 gene in colorectal carcinoma tissue and analyse its clinical significance.Methods Expression and distribution of PTEN and P53 protein were detected by S-ABC immunohistochemistry method in 59 colorectal carcinomas and 28 benign lesions on colorectal tissue.Results PTEN and P53 gene expression in colorectal cancer tissue of low differentiation group significantly up-regulated higher than the middle,high groups.Also significantly higher than the colorectal benign lesions and precancerous lesions groups.PTEN and P53 protein(AOD)were 0.512±0.016 and 0.326±0.028;in benign lesions of colorectal,AOD were 0.438±0.047 and 0.251±0.018.They were difference in statistically significant(P<0.05).Positive expression of PTEN and P53 gene has no relations with the patient's age,sexuality,tumor DUCKs stage,but they were co-related with the differentiation degree of malignant tumor andclinicalstage.Conclusion The expression of PTEN and p53 gene is related to the degree of differentiation of colorectal cancer.The lower degree of differentiation,the higher expression of PTEN and p53 gene.PTEN and p53 gene can be used as an important indicator of metastasis and prognosis of colorectal cancer.
