摘 要:目的:比较山莴苣素、山莴苣苦素对FFA诱导的HepG2细胞脂质代谢的影响。方法:采用山莴苣素和山莴苣苦素预处理FFA诱导的HepG2细胞模型48h,油红O观察细胞内脂滴,检测各组细胞内甘油三酯(TG)含量;比较脂质代谢相关因子(FABP1、HADHA、PPARγ、CPT1A、PPARα、PGC1α)与山莴苣素、山莴苣苦素的分子对接亲和力;采用实时荧光定量PCR法检测脂质代谢相关基因(FABP1、HADHA、PPARγ、CPT1A、PPARα、PGC1α)的mRNA相对表达量。结果:山莴苣素和山莴苣苦素均能抑制FFA诱导的HepG2细胞脂质堆积,降低细胞内脂滴数量,但山莴苣苦素对细胞内TG水平的影响没有山莴苣素明显;分子对接和荧光实时定量PCR结果表明,山莴苣苦素与6个脂代谢因子结合能均优于山莴苣素,山莴苣素、山莴苣苦素也均可上调PPARα、PPARγ、PGC1α、CPT1A、HADHA,下调FABP1的mRNA相对表达量,山莴苣苦素促进脂质代谢的能力也明显优于山莴苣素。结论:山莴苣素和山莴苣苦素均具有促进细胞内脂质代谢的能力,山莴苣苦素对于这6个脂质代谢基因的调控作用较山莴苣素更优。Objective:To compare the effects of lactucin and lactucopicrin on FFA-induced lipid metabolism in HepG2 cells.Methods:The FFA-induced HepG2 cell model was pretreated with lactucin and lactucopicrin for 48 h.Intracellular lipid droplets were observed by oil red O and intracellular triglyceride(TG)content was measured in each group.Comparison of the molecular docking affinities of lipid metabolism-related factors(FABP1,HADHA,PPARγ,CPT1A,PPARα,PGC1α)with lactucin and lactucopicrin.The relative mRNA expression of lipid metabolism-related genes(FABP1,HADHA,PPARγ,CPT1A,PPARα,PGC1α)was measured by real-time fluorescence quantitative PCR.Results:Both lactucin and lactucopicrin inhibited FFA-induced lipid accumulation in HepG2 cells and reduced the number of intracellular lipid droplets,but the effect of lactucopicrin on intracellular TG levels was not as pronounced as that of lactucin.Molecular docking and fluorescence real-time quantitative PCR results showed that all six lipid metabolism factors were better bound by lactucopicrin than lactucin,and both lactucin and lactucopicrin also up-regulated PPARα,PPARγ,PGC1α,CPT1A,HADHA,and down-regulated the relative mRNA expression of FABP1,and the ability of Lactucopicrin to promote lipid metabolism was also significantly better than that of lactucin.Conclusion:Both lactucin and lactucopicrin have the ability to promote intracellular lipid metabolism,the regulation of these six lipid metabolism genes by lactucopicrin was better than that by lactucin.
