摘 要:纤维化是多种疾病共同的病理环节,可影响任何器官,常导致组织器官实质细胞减少和结缔组织增生,引起进行性组织结构破坏和器官功能障碍[1-2]。纤维化是一个动态的、具有可逆性的病理过程,涉及多种细胞与信号转导间交联。转录激活因子(activating transcription factors,ATFs)是信号转导网络的关键枢纽,串联多种细胞信号,涉及内质网应激(endoplasmic reticulum stress,ERS)与靶细胞氧化应激、细胞增殖/凋亡、线粒体损伤及自噬等多种病理过程,可调节细胞功能,并影响疾病转归。目前对于ATFs在器官纤维化疾病中的表达和功能的报道较为零散,其作用机制尚不明确,本文将对ATFs的分类与特征,与器官纤维化的相关作用机制进行综述。Organ fibrosis was a common pathological change in a variety of chronic diseases,which was characterized by the increase of fibrous tissue and the decrease of parenchymal cells in tissues.Organ failure caused by organ fibrosis in the kidney,heart,lungs,liver,and other parenchymal organs raised the death and disability rates of patients.Currently,organ fibrosis incidence rates and associated medical costs have been rising yearly.Activating transcription factors(ATFs)were engaged in a variety of pathogenic processes,including as endoplasmic reticulum stress,mitochondrial damage,oxidative stress,cell proliferation,apoptosis,and autophagy,which were crucial in regulating cell function and disease development.Organ fibrosis has been shown to be closely associated to the activation of ATFs.As a result,blocking the aberrant activation of ATFs might be a possible therapeutic target for treating organ fibrosis.This article summarized the classification of ATFs and the research progress of ATFs involved in the signal pathways related to multiple organ fibrosis.
