摘 要:为了解河北省唐山市规模化奶牛场中牛病毒性腹泻病毒(BVDV)流行毒株的基因特征,本试验于唐山市2个规模化奶牛场采集了49份临床样品进行逆转录聚合酶链式反应(RT-PCR)检测,并对阳性样品进行病毒的分离,最终成功分离到1株非致细胞病变的BVDV毒株,命名为HB-1株。电镜观察结果显示,HB-1株具有典型的BVDV病毒粒子形态。对HB-1株全基因组序列进行扩增和测序,基因遗传进化分析结果显示,HB-1株属于BVDV-1m亚型。HB-1株结构蛋白E2全长序列糖基化位点和抗原表位预测结果显示,HB-1株E2蛋白含有4个保守的糖基化位点,其中抗原表位1高度保守。本试验为河北省进一步开展牛病毒性腹泻(BVD)流行病学调查、疫苗株筛选以及检测方法建立提供数据支撑。In order to assess the genetic characteristics of bovine viral diarrhea virus(BVDV)epidemic strains in large-scale dairy farms in Tangshan,Hebei province,this study collected 49 clinical samples from two large-scale dairy farms in Tangshan for reverse transcription-polymerase chain reaction(RT-PCR)detection,leading to the isolation of a non-cytopathic BVDV strain,named HB-1.The morphology of BVDV virus particles was observed by electron microscope,the whole genome sequence of HB-1 strain was amplified and analyzed,and the glycosylation and antigenic epitopes of HB-1 strain E2 protein sequence were predicted.The electron microscope results revealed that HB-1 strain had typical morphology of BVDV;the whole genome sequence analysis showed that HB-1 strain belonged to the subtype of BVDV-1m;the E2 protein sequence analysis indicated that HB-1 strain E2 protein contained four conserved glycosylation sites,and its epitope 1 was highly conserved.These results provide data support for the further epidemiological investigation of BVD,the screening of vaccine strains and the establishment of detection methods in Hebei province.
