摘　　要：目的探讨Teneurin跨膜蛋白3(TENM3)在胃癌中的表达及其对临床病理特征和预后的影响。方法利用癌症基因组图谱数据库(TCGA)数据库和基因型-组织表达(GTEx)数据库中胃癌及正常组织的转录组及临床数据分析TENM3 mRNA在胃癌样本中的表达及其与患者临床病理特征及预后的关系。通过R软件进行整理和分析数据,利用Wilcoxon秩和检验比较TENM3在肿瘤组织及正常组织中的表达情况,采用χ^(2)检验分析TENM3表达与临床病理指标之间的相关性,生存分析采用Cox回归分析。通过基因集富集分析(GSEA)探讨TENM3参与胃癌发生发展的可能机制。结果胃癌组织中TENM3的表达高于正常组织(1.062±0.927比0.649±0.663,P<0.001)。TENM3在T3~4期胃癌组织中的表达高于T1~2期胃癌组织(1.367±1.050比1.052±0.983,P<0.01);低分化胃癌组织中的表达高于中高分化组织(1.405±1.028比1.111±1.032,P<0.001)。TENM3高表达与患者不良预后有关(HR=1.165,P<0.05)。基因集富集分析结果表明TENM3可能通过一些代谢通路参与胃癌发生发展。结论TENM3在胃癌中发挥促癌作用,可作为胃癌患者的独立预后因素,有望成为治疗新靶标。Objective To investigate the expression of teneurin transmembrane protein 3(TENM3)in gastric cancer and its effect on clinicopathological features and prognosis.Methods The expression of TENM3 mRNA in gastric cancer samples and its relationship with clinicopathological characteristics and prognosis of patients were studied.The transcriptome and clinical data of gastric cancer and normal tissues in The Cancer Genome Atlas(TCGA)database and The Genotype-Tissue Expression(GTEx)database were used.R software was used to analyze the data,the Wilcoxon rank-sum test was used to compare the expression of TENM3 in tumor tissue and normal tissue,and the chi-square test was used to analyze the correlation between TENM3 expression and clinicopathological indicators,and survival analysis was analyzed by Cox regression analysis.Gene set enrichment analysis(GSEA)was used to explore the possible mechanism of TENM3 involved in the development of gastric cancer.Results The expression of TENM3 in gastric cancer tissues was higher than that in normal tissues(1.062±0.927 vs.0.649±0.663,P<0.001).The expression of TENM3 in T3-4 gastric cancer tissues was higher than that in T1-2 gastric cancer tissues(1.367±1.050 vs.1.052±0.983,P<0.01).The expression of TENM3 in poorly differentiated gastric cancer tissues was higher than that in moderately and well differentiated tissues(1.405±1.028 vs.1.111±1.032,P<0.001).High expression of TENM3 was associated with poor prognosis of patients(HR=1.165,P<0.05).The gene set enrichment analysis results indicated that TENM3 may be involved in the occurrence and development of gastric cancer through some metabolic pathways.Conclusion TENM3 plays a role in the development of gastric cancer.It can be used as an independent prognostic factor for gastric cancer and is expected to become a new target for treatment.
