皮肤鳞状细胞癌患者中STAT3、NF-κB p65表达及与其分化程度的关系研究

摘  要:目的探讨皮肤鳞状细胞癌(cSCC)患者信号传导与转录活化因子3(STAT3)、核因子-κB(NF-κB)p65表达与分化程度的关系。方法选取行手术治疗的58例cSCC患者,取手术切除cSCC组织标本作为研究组,距离肿瘤边缘5 cm以上的正常皮肤组织为对照组。采用免疫组化法检测STAT3、NF-κB p65表达情况,分析两者阳性表达与cSCC临床病理特征的关系。结果研究组STAT3、NF-κB p65阳性表达率较对照组高,差异有统计学意义(P<0.05);有淋巴转移者STAT3、NF-κB p65阳性表达率高于无淋巴转移者,Ⅲ~Ⅳ期STAT3、NF-κB p65阳性表达率高于Ⅰ~Ⅱ期者,且分化程度越低者STAT3、NF-κB p65阳性表达率越高,且肿瘤最大直径≥4 cm者STAT3、NF-κB p65阳性表达率高于<4 cm者,差异有统计学意义(P<0.05);STAT3、NF-κB p65阳性表达率与cSCC患者年龄、性别无关(P>0.05);STAT3、NF-κB p65阳性表达率与肿瘤最大直径、临床分期呈负相关(P<0.05),与分化程度、淋巴结转移呈正相关(P<0.05)。结论STAT3、NF-κB p65在cSCC中呈高表达,其阳性表达率与肿瘤最大直径、临床分期、分化程度、淋巴结转移密切相关,可作为判断患者病情和治疗的新靶点。Objective To investigate the relationship between the signal transduction,activator of transcription 3(STAT3)and nuclear factor-κB(NF-κB)P65 expression and differentiation degree in patients with cutaneous squamous cell carcinoma(cSCC).Methods 58 patients with cSCC who underwent surgical treatment were selected,and the cSCC tissue specimens from surgical resection were selected as the study group,and the normal skin tissue more than 5 cm from the tumor edge was selected as the control group.The expression of STAT3 and NF-κB p65 was detected by immunohistochemistry,and the relationship between the positive expression of STAT3 and NF-κB p65 and the clinicopathological characteristics of cSCC was analyzed.Results The positive expression rate of STAT3 and NF-κB P65 in the study group was higher than that in the control group,and the difference was statistically significant(P<0.05).The positive expression rates of STAT3 and NF-κB p65 in patients with lymphatic metastasis were higher than those without lymphatic metastasis,the positive expression rates of STAT3 and NF-κB p65 in patients with stageⅢ~Ⅳwere higher than those in patients with stageⅠ~Ⅱ,and the lower the differentiation degree was,the higher the positive expression rates of STAT3 and NF-κB p65 were,and the positive expression rates of STAT3 and NF-κB p65 in patients with tumor largest diameter≥4 cm were higher than those with tumor diameter<4 cm,and the difference was statistically significant(P<0.05).The positive expression rates of STAT3 and NF-κB P65 were not related to the age and gender of cSCC patients,and the difference was not statistically significant(P>0.05).The positive expression rates of STAT3 and NF-κB P65 were negatively correlated with the largest tumor diameter and clinical stage(P<0.05),but positively correlated with the degree of differentiation and lymph node metastasis(P<0.05).Conclusion STAT3 and NF-κB P65 are highly expressed in cSCC,and their positive expression rate is closely related to tumor maximum diameter,c
