摘 要:目的分析微RNA-466(miR-466)、微RNA-496(miR-496)、微RNA-153(miR-153)在子宫颈癌演进过程中的表达变化及意义。方法抽取2021年1月至2022年1月南阳市中心医院收治的子宫颈癌患者88例作为研究对象,采用荧光定量聚合酶链式反应(PCR)法检测所有研究对象的miR-496、miR-466、miR-153表达量;采用免疫组化法检测癌旁组织与癌组织中miR-466、miR-496、miR-153的表达情况。结果与癌旁组织相比,癌组织miR-466、miR-496表达量均升高,miR-153表达量下降(P<0.05)。miR-466、miR-496、miR-153表达量与子宫颈癌患者年龄无关(P>0.05),与子宫颈癌患者有无淋巴结转移、浸润深度、TNM分期、分化程度相关(P<0.05)。与无淋巴结转移、非肌层浸润、高分化、Ⅰ~Ⅱ期患者相比,肌层浸润、有淋巴结转移、中低分化、Ⅲ~Ⅳ期患者miR-153降低,miR-466、miR-496升高,差异有统计学意义(P<0.05)。miR-466、miR-496、miR-153之间相关性分析结果显示,miR-466与miR-496呈正相关(r=0.01,P=0.002),与miR-153呈负相关(r=-0.06,P=0.004);miR-496与miR-153呈负相关(r=-0.06,P=0.003)。结论 miR-466、miR-496在子宫颈癌组织中异常高表达,miR-153呈现低表达,且与患者浸润深度、有无淋巴结转移、分化程度、TNM分期相关。Objective To analyze the expression changes and significance of microRNA-466(miR-466),microRNA-496(miR-496),microRNA-153(miR-153)in evolution of cervical cancer.Methods Eighty-eight cervical cancer patients admitted to Nanyang Central Hospital from January 2021 to January 2022 were selected as research objects.The expression levels of miR-496,miR-466 and miR-153 in all subjects were detected by fluorescence quantitative polymerase chain reaction(PCR),while the expression level of miR-466,miR-496 and miR-153 in adjacent tissues and cancer tissues were detected by immunohistochemistry.Results Compared with adjacent tissues,the expression of miR-466 and miR-496 in cancer tissues increased,and expression of miR-153 in cancer tissues decreased(P<0.05).The expression of miR-466,miR-496 and miR-153 were not related to the age of cervical cancer patients(P>0.05),but were related to lymph node metastasis,invasion depth,tumor node metastasis(TNM)classification and differentiation degree of cervical cancer patients(P<0.05).Compared with patients without lymph node metastasis,non muscle layer invasion,highly differentiated,and stageⅠ-Ⅱ,expression of miR-153 decreased,while expression of miR-466 and miR-496 increased in the patients with muscle layer invasion,lymph node metastasis,medium and low differentiation and stageⅢ~Ⅳ(P<0.05).The results of correlation analysis among miR-466,miR-496 and miR-153 showed that miR-466 was positively correlated with miR-496(r=0.01,P=0.002),and was negatively correlated with miR-153(r=-0.06,P=0.004);there was a negative correlation between miR-496 and miR-153(r=-0.06,P=0.003).Conclusions The miR-466 and miR-496 are abnormally highly expressed in cervical cancer tissues,and the miR-153 shows low expression.It is associated with the depth of infiltration,metastasis of lymph node,degree of differentiation,and TNM stage.
