芥子油苷对豌豆蚜3种解毒酶活性的影响

摘  要:为明确植物次生物质芥子油苷对豌豆蚜(Acyrthosiphon pisum)体内3种解毒酶的影响,本研究采用豌豆蚜纯人工饲料薄膜饲养技术,设置对照组(0,CK)和3个不同浓度(0.94、1.87、2.86μmol·g^(-1))芥子油苷的处理组,测定豌豆蚜取食含有不同浓度芥子油苷的人工饲料后,体内3种解毒酶活性及可溶性蛋白含量的变化。结果表明:两种色型豌豆蚜取食添加不同浓度芥子油苷的人工饲料后,羧酸酯酶(CarE)、谷胱甘肽S-转移酶(GSTs)和细胞色素P450s酶活性逐渐升高,可溶性蛋白含量呈不同程度上升趋势;随着豌豆蚜取食时间延长和芥子油苷浓度升高,3种解毒酶活性逐渐升高;在芥子油苷浓度为0.94、1.87和2.86μmol·g^(-1)时,3种酶活性基本与对照之间差异显著(P<0.05)。研究结果表明,CarE、GSTs和P450s酶活性逐渐升高,三者相比,P450s酶活性显著高于CarE和GSTs活性(P<0.05),且在芥子油苷浓度为2.86μmol·g^(-1)处理下两种色型豌豆蚜的P450s酶活性分别是对照的4.60倍和1.90倍,该结果体现了两种色型豌豆蚜解毒酶对芥子油苷的响应机制不同。The aim of this study was to investigate the effects of plant secondary glucosinolates on three detoxification enzymes in aphid pea.In this study,pea aphid was fed pure artificial diet;the treatment groups include the control group(0μmol·g^(-1))and three groups fed with diet supplemented with glucosinolates at different concentrations(0.94,1.87,2.86μmol·g^(-1)).Changes in detoxifying enzyme activity and soluble protein content in artificial diets containing different glucosinolate concentrations were measured.The results showed that the enzyme activities of carboxylesterase(CarE),glutathione S-transferase(GSTs),and cytochrome P450s(P450s)increased gradually when two morphs of pea aphid were fed artificial diets supplemented with different concentrations of glucosinolates,and the soluble protein content increased in varying degrees.With increases in the feeding time and glucosinolate concentration,the activities of the three detoxification enzymes increased.Compared to 0.94μmol·g^(-1)group,the 1.87 and 2.86μmol·g^(-1)groups were significantly different(P<0.05).In this study,the enzyme activities of CarE,GSTs,and P450s gradually increased.Compared to CarE and GSTs,the enzyme activities of P450s were significantly higher(P<0.05),and at 2.86μmol·g^(-1),the enzyme activity of P450s was 4.60 and 1.90 times higher than that of the control,respectively.The results indicated that the detoxification metabolism of different pea aphids had different response mechanisms to glucosinolates.
