摘  要:目的:基于网络药理学探讨橘核活精方治疗男性不育症的分子作用机制。方法:应用中药分子机制的生物信息学分析工具(BATMAN-TCM)收集橘核活精方的有效成分,并预测其作用靶点;通过GeneCards、DrugBank、CTD数据库获取治疗男性不育症的作用靶点;利用MLC在线工具将筛选后的作用靶点相互交集以获取药物和疾病共有靶点基因。利用String平台对共有基因构建蛋白质-蛋白质相互作用网络;利用Cytoscape软件(v3.9.0)构建“有效成分-靶点”网络图;利用Hiplot可视化平台对共有基因进行基因本体论(GO)功能富集分析、京都基因与基因组百科全书(KEGG)的生物通路富集分析。结果:筛选后得到橘核活精方中作用于男性不育症的有效成分共有152种,橘核活精方治疗男性不育症的潜在靶点共126个,涉及磷脂酰肌醇-3激酶(Phosphatidylinositol 3-kinase,PI3K)-丝氨酸/苏氨酸激酶(Akt)、缺氧诱导因子-1(Hypoxia Inducible Factor-1,HIF-1)等信号通路,相关通路涉及药物、肽激素刺激、氧化应激、活性氧合成与代谢、脂质代谢等因素。结论:橘核活精方治疗男性不育症具有多成分、多靶点、多通路的特点,体现了中药复方的整体性。本研究期待为进一步探索橘核活精方分子作用机制提供新思路。Objective:To explore the molecular mechanism of action of the Juhe Huojing prescription(橘核活精方)based on the network pharmacology.Methods:The effective components were collected and the possible targets were predicted by bioinformatics analysis tool for molecular mechanism of traditional Chinese medicine(BATMAN-TCM),the contributing targets of male infertility were obtained from several databases such as GeneCards,DrugBank,CTD database,the predicted targets were intersected with by MLC to screen out the common targets of prescription and disease.The protein-protein interaction network for common genes was constructed by String platform.The ingredient-target network was established to clarify the active compounds and potential targets by Cytoscape(v3.9.0).The gene ontology(GO)and Kyoto encyclopedia of genes and genomes(KEGG)enrichment analyses were carried out by Hiplot,the data visualization platform.Results:Totally 152 active components and 126 disease-related targets with high correlation with male infertility were observed after being screened,which were mainly involved in signaling pathways including PI3K-Akt,HIF-1,and correlation paths including drug,peptide hormone stimulation,oxidative stress,reactive oxygen species synthesis and metabolism,and lipid metabolism.Conclusion:The Juhe Huojing prescription has the characteristics of multiple components,targets,pathways in the treatment of male infertility which reflected the overall feature of TCM compound prescription,thereby providing new directions and clues for further research on the molecular mechanism of action of the Juhe Huojing prescription.
