摘  要:大球盖菇是我国近年来广泛栽培的一种食用菌,具有较高的食用价值和经济价值。为了获得大球盖菇原生质体单核体,主要进行了不同品种大球盖菇原生质体制备及单核化试验,用血球计数板计数原生质体总数,挑取再生原生质体的单个菌落,25℃倒置黑暗恒温培养,倒置显微镜观察菌丝锁状联合情况,筛选获得大球盖菇单核体。结果表明:大球盖菇原生质体再生率均较高,‘土肥’品种再生率最高,达1.37%,‘高邮’品种再生率次之为1.13%,‘大T’品种的再生率最低为0.90%;单核化率‘大T’品种高达85.2%,‘土肥’的单核化率次之为66.0%,‘高邮’的单核化率最低为59.8%。对不同大球盖菇品种的单核体表观形态特性进行观察分析表明:利用大球盖菇原生质体技术选育新品种提供单核体,可丰富食用菌原生质体单核体资源,为研究遗传及育种提供材料。Stropharia rugosoannulata is a kind of edible mushroom widely cultivated in China in recent years,which has high edible value and economic value.In order to obtain the protoplast mononuclear of S.rugosoannulata.In this paper,the different varieties of S.rugosoannulata were tested on the preparing protoplast and protoplast monokarygenesis.Hemocytometer counted the amount of protoplast,then get the single colony form the protoplast regenration,cultivate inversion in 25℃in darkness,inverted microscope observed clamp connection of mycelium,mononuclears were obtained by screening.The result showed that the protoplast regeneration rate of S.rugoso-annulata was high.Three varieties of‘Tufei’regeneration rate was the highest,up to 1.37%,‘Gaoyou’regeneration rate was 1.13%and‘T’variety regeneration rate was the lowest,was 0.90%.Monokaryotization rate of‘T’variety as high as 85.2%,and monokaryotization rate of‘Tufei’was 66%,‘Gaoyou’monokaryotization rate as low as 59.8%.By observing and analyzing the morphological characteristics of mononuclears in different varieties of S.rugosoannulata,we can provide mononucleotide for breeding new varieties by using the protoplast technology of S.rugosoannulata,which can enrich the resources of protoplast mononuclear,and provide raw materials for research on genetics and breeding.
