摘 要:目的:优化回流-酶法、超声-酶法提取鸡冠花总黄酮的工艺条件。方法:聚酰胺柱纯化样品后于258 nm处测定总黄酮含量。以鸡冠花总黄酮提取率为指标,用单因素试验法优化酶种类、加酶量、pH、温度和酶解时间等酶解条件,用正交试验法优化提取溶剂、次数和时间等回流-酶法、超声-酶法的提取条件。结果:加3倍量乙酸-乙酸钠缓冲液(pH 4)到鸡冠花粉末中,再加纤维素酶、果胶酶(1∶50),搅拌,40℃下保温60 min,再用60%乙醇按如下方法提取总黄酮:回流提取3次(8、6、6倍量溶剂。第1次提取时加5倍量95%乙醇,混匀得8倍量60%乙醇),每次回流30 min,总黄酮提取率95.0%;超声提取2次(12、8倍量溶剂),每次提取20 min,总黄酮提取率94.8%。结论:优化得出的超声-酶法提取工艺高效、节能、简便、环保,可为开发利用鸡冠花总黄酮提供实验基础。Objective:To optimize the technological conditions of enzyme-assisted reflux and ultrasonic-wave extractions for total flavonoids from Celosia cristata.Methods:The total flavonoids from C.cristata purified with polyamide column were determined at 258 nm.With the extraction rate of total flavonoids from C.cristata as index,the technological conditions of enzyme hydrolysis,including the kind and dosage of enzyme,pH,temperature and time,were optimized by single factor test;and the technological conditions of enzyme-assisted reflux and ultrasonic-wave extractions,including solvent,frequency and time,were optimized by orthogonal test.Results:C.cristata powder was pretreated with cellulase(1∶50)and pectinase(1∶50)for 60 min in 3 times volume of acetic acid-sodium acetate buffer(pH 4)at 40℃,and then total flavonoids were extracted with 60%ethanol by following methods:extracted 3 times under reflux for 30 min each time(8,6,6 times volume solvent.Eight times volume 60%ethanol was prepared by adding 5 times volume of 95%ethanol at the first time),and the extraction rate of total flavonoids was 95.0%;extracted 2 times with ultrasonic-wave for 20 min each time(12,8 times volume solvent),and the extraction rate of total flavonoids was 94.8%.Conclusion:The optimum process of enzyme-assisted ultrasonic-wave extraction is efficient,energy-saving,simple and environmentally friendly,and provides an experimental base for development and application of total flavonoids from C.cristata.
