摘 要:目的:研究橘皮素对双氧水致心肌细胞氧化损伤的保护作用及与PI3K/Akt(酰肌醇3-激酶/蛋白激酶B)信号通路的相关性。方法:培养H9C2心肌细胞,分3组,分别为:正常组(Control)、氧化损伤组(Oxidative Damage Group,ODG)、橘皮素组(Tangeretin);其中氧化损伤组H9C2心肌细胞通过双氧水构建氧化损伤模型,橘皮素组在氧化损伤组的基础上使用橘皮素预处理;通过CCK8实验测定各组细胞的活力值,流式细胞术检测各组细胞内ROS(活性氧)的表达情况,Western blot检测各组PI3K/AKT信号通路相关蛋白的表达。结果:与Control组相比,ODG组与Tangeretin组细胞活力值均明显下降(均P<0.05),细胞内ROS表达量均明显上升(均P<0.05),ODG组细胞p-Akt/Akt的表达明显下降(P<0.05);与ODG组相比,Tangeretin预处理后,细胞活力值明显提升(P<0.05),细胞内ROS表达量明显降低(P<0.05),PI3K与p-Akt/Akt的表达明显上调(均P<0.05)。结论:橘皮素预处理可有效发挥双氧水致心肌细胞氧化损伤的保护作用,其机制可能与PI3K/AKT信号通路的激活相关。Objective:To research the protective effect of Tangeretin on oxidative damage of cardiomyocytes induced by hydrogen peroxide and its correlation with PI3K/Akt signaling pathway.Methods:H9C2 cardiomyocytes were cultured and divided into three groups:control group,oxidative damage group(ODG)and Tangeetin group;H9C2 myocardial cells in the oxidative damage group were cultured with hydrogen peroxide to establish an oxidative damage model,and the Tangeretin group was pre-cultured with Tangeretin on the basis of the oxidative damage group;The optimal protective concentration of Tangeretin against oxidative damage was determined by CCK8 experiment,the expression of ROS in each group was detected by flow cytometry,and the expressions of PI3K/Akt signal pathway related proteins in each group were detected by Western blot.Results:Compared with the control group,the cell viability of ODG group and Tangeetin group were decreased significantly(all P<0.05),and the intracellular ROS expression was increased significantly(all P<0.05),p-Akt/Akt expression in ODG group was lowered(P<0.05).Compared with ODG group,the cell viability in Tangeetin group was increased significantly(P<0.05),the intracellular ROS expression was decreased significantly(P<0.05),and the expressions of PI3K and p-Akt/Akt were increased significantly(all P<0.05).Conclusion:Tangeetin pretreatment can effectively protect myocardial cells from oxidative damage induced by hydrogen peroxide,and its mechanism may be related to the activation of PI3K/Akt signaling pathway.
