摘 要:【目的】介绍一种利用数码显微镜进行细胞计数的简便方法。【方法】体外分离培养猪肺微血管内皮细胞(microvascular endothelial cells,MVECs)和猪外周血中性粒细胞,利用数码显微镜对不同密度的2种细胞进行计数,并与利用自动细胞计数仪和血细胞计数板的计数结果比较,分析其计数结果准确性、计数效率和绘制细胞生长曲线情况。【结果】计数不同密度水平的细胞时,数码显微镜计数法与细胞计数仪法的结果均无显著性差异,对MVECs计数结果的变异系数低于中性粒细胞;计数MVECs用时少于细胞计数板法和细胞计数仪法,计数中性粒细胞时用时多于另外2种方法;3种方法绘制的细胞生长曲线呈趋势一致性,数码显微镜计数法与细胞计数板法、细胞计数仪法的相关系数分别为0.994和0.990。【结论】数码显微镜细胞计数法是一种便捷、易操作、准确、省时且不增加试验成本的计数方法,尤其适合于贴壁型细胞的原位计数,值得在细胞试验中推广应用。[Objective]To introduce a simple method for cell counting using the digital microscope(DM).[Methods]Porcine lung microvascular endothelial cells(MVECs)and neutrophils were separated and cultured in vitro.They were counted using the DM,and their accuracy,counting times and the growth curve plotting were compared to those from the cell counting chamber(CCC)and the cell counter(CC).[Results]When counting the cells at different densities,there were no significant difference between the results using the DM and CC,and the variable coefficients of MVECs were lower than that of neutrophils.The time counting MVECs using the DM was shorter than that using the CCC or CC,while the time counting neutrophils using the DM was more than that using the CCC or CC.Moreover,the growth curves of MVECs from three methods were in agreement,and the correlation coefficients between the DM and CCC and CC were 0.994 and 0.990,respectively.[Conclusion]The DM counting method is convenient,maneuverable,accurate,time-saving,and costless.It is especially suitable for counting adherent cells,and deserves to be popularized in cell experiments.
