摘 要:目的 探讨FDX1基因在乳腺癌中的表达和预后价值,分析其潜在的生物学功能及对免疫微环境的影响。方法 利用TCGA数据库获取乳腺癌样本数据信息,分析FDX1在肿瘤中的的表达情况;免疫组化法检测FDX1在乳腺组织中的蛋白表达;Bc-GeExminer 3.0数据库分析在乳腺癌中FDX1表达与ER、PR、HER2表达的关系;Kaplan-Meier Plotter分析FDX1表达与总生存期(OS)、无复发生存期(RFS)、无远处转移生存期(DMFS)相关性;采用Timer数据库和GSVA数据库分析乳腺癌中FDX1表达与免疫细胞浸润的相关性;利用LinkedOmic中基因集富集分析(GSEA)探讨FDX1介导的生物学功能机制。结果 在TCGA数据集中,FDX1在乳腺癌肿瘤组织中表达降低;免疫组化结果显示,FDX1在乳腺癌肿瘤组织中蛋白水平降低;FDX1表达与PR表达、ER表达、PAM50状态相关(P<0.001),与年龄、T分级、N分级、M分级、病理分级、组织学分型、解剖肿瘤部位、是否接受放射治疗均无关(P>0.05);Bc-GeExminer 3.0数据库免疫组化结果提示FDX1表达与ER、PR、HER2表达密切相关;FDX1高表达乳腺癌患者其OS、RFS、DMFS预后较好相关(P<0.05);FDX1表达与乳腺癌肿瘤免疫细胞浸润显著相关(P<0.05);GSEA结果显示,FDX1表达与核糖体、氧化磷酸化、帕金森病密切相关。结论 乳腺癌组织中FDX1的表达异常,低表达患者的预后不良,FDX1可能通过肿瘤免疫浸润细胞改变肿瘤微环境,促进乳腺癌的发生发展,可作为潜在评估乳腺癌预后的分子标志物。Objective To investigate the expression of FDX1 and clarify the clinical significance and prognostic value of FDX1 in breast cancer,as well as its potential biological function and effect on the microenvironment.Methods FDX1 expression and its clinical information were obtained from TCGA datasets.Protein expression levels of FDX1 in breast tissue were detected by immunohistochemistry.The Bc-GeExminer 3.0 database was used to analyze the relation between FDX1 and ER,PR and HER2 expression in breast cancer.Kaplan-Meier Plotter was used to analyze the correlation between FDX1 expression and overall survival(OS),relapse-free survival(RFS)and distant metastasis free survival(DMFS).The asscioation between FDX1 expression and immune cell infiltration in breast cancer was investigated using the Timer and GSVA database.Gene Set Enrichment Analysis(GSEA)in LinkedOmic was used to explore the biological function mediated by FDX1.Results In the TCGA dataset,the expression of FDX1 was decreased in breast cancer tissues.Inmunohistochemical result showed that the protein level of FDX1 decreased in breast cancer tumor tissue.FDX1 expression was correlated with PR status,ER status,PAM50 status(P<0.001),but not related to T/N/M stage,pathologic stage,histology classification,anatomic neoplasm subdivisions,whether receiving radiation therapy(P>0.05).The immunohistochemical results of Bc-GeExminer 3.0 database indicated that the expression of FDX1 was closely related to the expression of ER,PR and HER2.High expression of FDX1 was closely related to better prognostic in the OS,RFS and DMFS of BC patients(P<0.05).The expression of FDX1 was closely related to immune infiltration.GSEA results showed that FDX1 expression was associated with ribosome,oxidative phosphorylation,Parkinson’s disease(P<0.05).Conclusion The expression of FDX1 in breast cancer tissue is abnormal,and patients with low expression have poor prognosis.FDX1 may change the tumor microenvironment through affect tumor immune infiltrating cells and promote the occurrenc
