骨保护素对大鼠脑出血后iNOS表达影响的研究

摘　　要：目的探讨骨保护素对大鼠脑出血后iNOS表达的影响。方法将SD大鼠随机分为Sham组、ICH组、OPG组,每组又分为12 h、24 h、72 h共3个亚组。在各个时间点进行Bederson评分,应用免疫组化技术观察不同时间点大鼠脑出血血肿周围iNOS的表达。结果各组大鼠12 h神经功能缺损最重,之后症状逐渐减轻。12 h ICH组、OPG组与Sham组相比,差异均具有统计学意义(P<0.05)。脑含水量比较,12 h、24 h、72 h,ICH组、OPG组与Sham相比差异有统计学意义(P<0.05);72 h OPG组与ICH组相比差异具有统计学意义(P<0.05)。各组血肿周围脑组织中iNOS的表达:12 h、24 h、72 h,OPG组、ICH组与Sham相比差异有统计学意义(P<0.05);72 h OPG组明显高于ICH组,两者相比差异具有统计学意义(P<0.05)。结论大鼠侧脑室注入重组OPG可引起血肿周围iNOS的表达显著增加,OPG可能增加脑出血后的炎性反应。Objective To investigate the effect of osteoprotegerin(OPG)on inducible nitric oxide synthase(iNOS)expression in rats with intracerebral hemorrhage(ICH).Methods Male Sprague-Dawley(SD)rats were divided into three groups:Sham group,ICH group,and OPG group.Each group was randomly divided into four subgroups:12 h,24 h and 72 h after operation.The Bederson score was used at various time after operation.The expression of iNOS in the cerebral tissue surrounding hematoma was determined by immunohistochemical technique.Results The behavioral scores:the neurological deficit scores were the most severe at 12 h after ICH in each group,and then the neurological symptoms gradually diminished.Compared with Sham groups,the Bederson score in ICH and OPG group was obviously increased,and showed statistically differences at 12 h after operation(P<0.05).The brain water content:the brain water content was significantly greater in ICH and OPG group compared with Sham group at 12 h,24 h and 72 h after operation.The expression of iNOS in the cerebral tissue surrounding hematoma:the expression of iNOS was significantly elevated in ICH and OPG group compared with Sham group at 12 h,24 h and 72 h after operation.The expression of iNOS in OPG group was notably higher than that in ICH group at 72 h(P<0.05).Conclusion Injection of recombinant OPG into the lateral ventricle of rat can increase the expression of iNOS around the hematoma.OPG might promote the inflammatory responses after ICH.
