血浆RNF180基因甲基化与胃癌风险相关性的Meta分析

摘　　要：目的系统评价血浆环指蛋白180(ring finger protein 180,RNF180)甲基化与胃癌及其不同临床病理特征的相关性。方法计算机检索The Cochrane Library、EMbase、PubMed等数据库,查找关于血浆RNF180甲基化与胃癌及其不同临床病理特征相关性分析的病例-对照研究,设定检索时限为从建库到2021年6月。采用RevMan5.3软件行Meta分析。结果纳入7个研究,共1104例患者,其中胃癌组551例,正常胃组织组553例。Meta分析结果显示(1)血浆RNF180甲基化在胃癌组与正常胃组织组[OR=5.04,95%CI(3.65,6.97),P<0.00001]的差异有统计学意义。(2)血浆RNF180甲基化在胃癌分化组与未分化组[OR=0.15,95%CI(0.07,0.33),P<0.00001]、伴淋巴结转移组与无淋巴结转移组[OR=3.47,95%CI(2.22,5.40),P<0.00001]、有远处转移组与无远处转移组[OR=5.78,95%CI(2.27,14.71),P<0.0002]、TNM临床分期Ⅰ-Ⅱ期组与Ⅲ-Ⅳ期组[OR=0.25,95%CI(0.16,0.39),P<0.00001]的差异有统计学意义;而在男组与女组[OR=0.79,95%CI(0.53,1.15),P=0.22]和≤60岁组与>60岁组[OR=0.84,95%CI(0.49,1.44),P=0.54]的差异均无统计学意义。结论血浆RNF180基因甲基化与胃癌有显著相关性。因受纳入研究的质量和数量制约,以上结论仍需更多高质量研究支撑。Objective To systematically evaluate the correlation between plasma RNF180 gene methylation and gastric cancer as well as its different clinicopathological features.Methods The Cochrane Library,EMbase,PubMed and other databases were electronically searched to collect the correlation between plasma RNF180 gene methylation and gastric cancer as well as its different clinicopathological features from inception to June 2021.Then meta-analysis was implemented by using RevMan5.3 software.Results A total of 7 case-control studies involving 1104 objects were considered,of which,551 cases in the gastric cancer group and 553 cases in the normal gastric tissue group.The results of meta-analysis showed that,as for plasma RNF180 gene methylation,significant differences were found in gastric cancer vs.normal gastric tissues(OR=5.04,95%CI 3.65 to 6.97,P<0.00001),and in gastric cancer tissues with differentiated vs.undifferentiated(OR=0.15,95%CI 0.07 to 0.33,P<0.00001),with vs.without lymph node metastasis(OR=3.47,95%CI 2.22 to 5.40,P<0.00001),with vs.without distant metastasis(OR=5.78,95%CI 2.27 to 14.71,P<0.0002),and TNMⅠ-Ⅱvs.Ⅲ-Ⅳ(OR=0.25,95%CI 0.16 to 0.39,P<0.00001).However,no significant difference was found in gastric cancer for male vs.female(OR=0.79,95%CI 0.53 to 1.15,P=0.22)and no less than vs.less than 60 years(OR=0.84,95%CI 0.49 to 1.44,P=0.54).Conclusions Plasma RNF180 gene methylation is significantly correlated with gastric cancer.Restricted by the quality and quantity of the included studies,the above conclusions still need to be supported by more high-quality studies.
