摘  要:目的:观察电针对去卵巢脂质代谢紊乱大鼠肝组织形态结构、雌二醇(E2)及雌激素受体(ER)α、ERβ表达的影响,对大鼠肝脏中雌激素相关差异基因进行生物信息学分析,为电针调节雌激素水平进而改善脂质代谢紊乱提供依据。方法:选用SD雌性大鼠55只,随机分为空白组、假手术组、模型组、西药组、电针组,各11只。空白组给予普通饲料喂养,其余各组均以高脂饲料喂养。电针组选穴足三里、内关、三阴交、肝俞、丰隆(双侧)治疗。治疗结束后,对肝脏组织进行HE和油红O染色分析;ELISA检测血清E2含量以及肝脏ERα、ERβ表达水平;运用DAVID数据库对肝脏组织差异表达基因进行富集分析,STRING数据库构建基因互作网络,对雌激素相关的差异表达基因进行可视化分析。结果:与空白组比较,模型组肝组织出现明显的结构破坏和脂肪堆积,血清E2含量减少(P<0.05),肝脏ERα表达降低(P<0.05)。与模型组比较,电针组和西药组肝组织结构破坏和脂肪堆积程度降低;电针组E_(2)含量增多,但差异无统计学意义;电针组ERα、ERβ表达水平差异无统计学意义。肝脏转录组测序结果提示,组间差异表达基因在雌激素信号通路等均有富集,并且雌激素信号通路相关的基因间彼此存在互作关联。结论:电针对去卵巢脂质代谢紊乱大鼠有较好的治疗效应,其可能是通过对雌激素及其受体以及雌激素信号通路相关基因表达的调控来实现的。Objective:To observe the effect of electroacupuncture on liver tissue morphology,estradiol(E2)and estrogen receptor(ER)α,ERβin ovariectomized rats with lipid metabolism disorder.The bioinformatics analysis of estrogen related differential genes in rat liver can provide the basis for electroacupuncture to regulate estrogen level and improve lipid metabolism disorder.Methods:A total of 55 female SD rats were randomly divided into blank group,sham operation group,model group,western medicine group and electroacupuncture group,with 1l rats in each group.The blank group was fed with ordinary diet,and the other groups were fed with high-fat diet.Electroacupuncture points Zusanli,Neiguan,Sanyinjiao,Ganshu,Fenglong(all acupoints were bilateral)were selected for treatment.After the treatment,HE and oil red staining were performed on the liver tissue;Serum E_(2) and liver ERα,ERβexpression level were detected by ELISA;DAVID database was used to enrich and analyze differentially expressed genes in liver tissue,and STRING database was used to construct gene interaction network to visually analyze estrogen related differentially expressed genes.Results:Compared with the blank group,the liver tissue of the model group showed obvious structural damage and fat accumulation,the serum E,content decreased(P<0.05),and the liver ERαexpression decreased(P<0.05).Compared with the model group,the damage of liver tissue structure and the degree of fat accumulation in the electroacupuncture group and the western medicine group were decreased,the E,content in the electroacupuncture group was increased,but there was no statistical significance,and the ERα,ERβexpression level was not statistically significant.The results of liver transcriptome sequencing showed that the differentially expressed genes were enriched in the estrogen signaling pathway,and the genes related to the estrogen signaling pathway interacted with each other.Conclusion:Electroacupuncture has a good therapeutic effect on ovariectomized rats with lipid metabolism di
