摘  要:[目的]探讨溃疡性结肠炎(UC)外周细胞连接蛋白(connexin,Cx)26及Cx43表达情况。[方法]选择8周龄雄性大鼠30只,按照随机数字表法将所有大鼠分为实验组(n=15)和对照组(n=15)。其中实验组大鼠采用2,4,6-三硝基苯磺酸(TNBS)/乙醇灌肠建立大鼠UC模型,而后分别取2组大鼠外周血、脾脏及肠道黏膜固有层细胞。使用蛋白质印迹分析检测外周血、脾脏及肠道黏膜固有层淋巴细胞中Cx26及Cx43表达情况,并对外周血中干扰素-γ(IFN-γ)、白细胞介素-4(IL-4)水平进行检测,分析2组之间差异。而后选择确诊UC的患者50例为UC组(n=50),并纳入同期体检的健康人群50例为健康组(n=50)。抽取外周静脉血通过蛋白质印迹分析检测外周血中Cx26及Cx43表达情况,并通过建立ROC曲线了解上述2个指标对于UC诊断的效能。[结果]实验组大鼠IFN-γ高于对照组,而IL-4则低于对照组,有统计学意义(P<0.001)。2组大鼠组内外周血、脾脏及肠道黏膜固有层单核细胞中Cx26及Cx43表达水平比较差异无统计学意义(P>0.05);实验组外周血、脾脏及肠道黏膜固有层单核细胞中Cx26及Cx43表达水平显著高于对照组(P<0.05)。2组大鼠的各部位单核细胞中Cx26及Cx43表达水平与IFN-γ呈显著正相关(P<0.001);2组大鼠的各部位单核细胞中Cx26及Cx43表达水平与IL-4呈显著负相关(P<0.001)。UC组Cx26及Cx43表达水平高于健康组,差异有统计学意义(P<0.001)。Cx26及Cx43以及二者联合检验对UC患者预测均具有一定价值(均P<0.05);联合检测的AUC显著高于Cx26及Cx43,差异有统计学意义(P<0.05)。[结论]UC患者外周血细胞Cx26及Cx43的表达水平显著升高,但将其作为UC临床诊断指标,尚需要进一步研究大样本予以证实。[Objective]To investigate the expression of Cx26and Cx43in peripheral cells from ulcerative colitis(UC)subjects.[Methods]Thirty male rats of 8weeks old were selected,and all rats were divided into experimental group(n=15)and control group(n=15)according to the random number table method.The experimental group of rats used 2,4,6-trinitrobenzenesulfonic acid(TNBS)/ethanol enema to establish a rat UC model,and then the peripheral blood,spleen and intestinal mucosal lamina propria cells of the two groups of rats were taken.Western blot analysis was used to detect the expression of Cx26and Cx43in peripheral blood,spleen and intestinal lamina propria lymphocytes,and the levels of IFN-γand IL-4in peripheral blood were detected to analyze the differences between the two groups.Then,50patients diagnosed with UC in our hospital from January 2020to May 2021were selected as the UC group(n=50),and 50 healthy people included in the physical examination during the same period were selected as the healthy group(n=50).Peripheral venous blood was drawn to detect the expression of Cx26and Cx43in peripheral blood by Western blot analysis,and the ROC curve was established to understand the effectiveness of the above two indicators in the diagnosis of UC.[Results]The IFN-γof the experimental group was higher than that of the control group,while the IL-4was lower than that of the control group.The difference was statistically significant(P<0.001).There was no significant difference in the expression of Cx26and Cx43in peripheral blood,spleen and lamina propria monocytes of the intestinal mucosa between the two groups of mice(P>0.05);the experimental group had no significant difference in the expression of monocytes in the lamina propria of the peripheral blood,spleen and intestinal mucosa.The expression levels of Cx26and Cx43were significantly higher than those of the control group(P<0.05).The expressions of Cx26and Cx43in monocytes in each part of the two groups of mice were significantly positively correlated with IFN-γ(P<0.001).Th
