摘 要:脂肪间充质干细胞(adipose-derived stem cells,ASCs)来源广泛,增殖能力强,成为近期实验和临床研究的热点。已有研究表明,ASCs可缓解神经病理性疼痛(neuropathic pain,NPP)。但其机制仍需进一步研究。本研究旨在探讨ASCs对谷氨酸(glutamate,Glu)诱导的SH-SY5Y细胞损伤模型的作用及其可能的机制。CCK-8检测结果显示,谷氨酸诱导的SH-SY5Y细胞增殖活力显著降低(P<0.05)。流式细胞术检测结果显示,谷氨酸诱导的SH-SY5Y细胞凋亡率显著升高(P<0.05)。ELISA检测结果显示,谷氨酸诱导的SH-SY5Y细胞中IL-1β、TNF-α和IL-6浓度显著升高(P<0.05)。Western印迹检测结果显示,谷氨酸诱导的SH-SY5Y细胞PI3K和AKT磷酸化水平、裂解胱天蛋白酶3(cleaved-Caspase-3)水平显著升高(P<0.05)。与Glu组相比,Glu+ASCs组SH-SY5Y细胞增殖活力显著升高(P<0.05),细胞凋亡率、细胞中IL-1β、TNF-α、IL-6浓度、PI3K和AKT磷酸化水平、cleaved-Caspase-3蛋白质水平显著降低(P<0.05)。总之,研究结果提示,ASCs对谷氨酸诱导的SH-SY5Y细胞损伤具有一定的保护作用,其作用机制可能与PI3K/AKT信号通路有关。Adipose-derived stem cells(ASCs)are widely available and have a high proliferative capacity,making them a recent hot topic for experimental and clinical research.ASCs have been shown to relieve neuropathic pain(NPP).However,the mechanism still needs further study.The aim of this study was to investigate the role and mechanisms of ASCs on SH-SY5Y cell injury model induced by glutamate(Glu).CCK-8 assay results showed that the proliferation viability of Glu-induced SH-SY5Y cells was significantly reduced(P<0.05).The results of flow cytometry assay showed that the apoptosis rate of Glu-induced SH-SY5Y cells was significantly increased(P<0.05).The levels of IL-1β,TNF-αand IL-6 in Glu-induced SH-SY5Y cells were significantly increased(P<0.05)via ELISA assay.The PI3K and AKT phosphorylation levels and cleaved-Caspase-3 protein levels were significantly elevated in Glu-induced SY5Y cells(P<0.05)by Western blot assay.Furthermore,compared with Glu group,the proliferation viability of SH-SY5Y cells was significantly increased(P<0.05),and the apoptosis rate,the levels of IL-1β,TNF-α,IL-6,the phosphorylation levels of PI3K and AKT l,and cleaved-Caspase-3 protein level in cells were significantly decreased(P<0.05)in Glu+ASCs group.In summary,our experimental results suggest that ASCs have a protective effect on Glu-induced SH-SY5Y cell injury,and the mechanism may be related to PI3K/AKT signaling pathway.
