摘 要:目的探讨全身性炎性反应中NLRP3(NOD-like receptor protein 3,NLRP3)炎症小体的作用。方法将8周龄雄性C57BL/6J小鼠随机分为对照组和手术组(每组7只),双侧颈动脉暴露手术后24 h收集小鼠血浆、肝脏、脾脏和脑组织,采用定量聚合酶链式反应法(qPCR)检测不同组织中白细胞介素(interleukin,IL)-1β和IL-18的mRNA表达水平,采用Western blot检测NLRP3蛋白表达水平,酶联免疫吸附法(ELISA)检测IL-1β和IL-18浓度。结果与对照组相比,手术组小鼠血浆IL-1β表达水平增高(P<0.01),肝脏和脾脏IL-1βmRNA、IL-18 mRNA和NLRP3蛋白也增加(P均<0.05)。在脑组织中,与对照组相比,手术组小鼠IL-1β和IL-18的mRNA水平均增高(P均<0.05)。ELISA法检测结果显示,手术组小鼠脑组织IL-1β和IL-18蛋白水平均高于对照组(P均<0.01)。Western blot结果显示,手术组NLRP3蛋白水平较对照组增多(P<0.05)。结论手术介导的全身性炎性反应与NLRP3炎症小体途径有关。Objective To investigate the role of NOD-like receptor protein 3(NLRP3)contributes to systemic inflammatory response.Methods Male C57BL/6J mice(8 weeks age)were randomly divided into the control group and the surgical group(7 mice/group).After 24 h of the bilateral carotid artery exposure surgery,specimens from plasma,liver,spleen,and brain were respectively collected.The mRNA expressions of IL-1βand IL-18 were separately measured by quantitative PCR(qPCR).Western blot was used to detect the expression of NLRP3.Enzyme-linked immunosorbent assay(ELISA)was used to determine the expressions of IL-1βand IL-18.Results Compared with the control group,the plasma level of IL-1βin the surgical group was significantly increased(P<0.01).IL-1βmRNA,IL-18 mRNA,and NLRP3 protein expression also increased in liver and spleen tissues(P all<0.05).In the brain tissue,the results of qPCR showed that the mRNA levels of IL-1βand IL-18 were higher in the operation group compared to the control group(P all<0.05).When compared to the control group,the surgery group’s brain tissue had greater amounts of IL-1βand IL-18 protein,according to the ELISA results(P all<0.01).Western blot results revealed that the level of NLRP3 protein was higher in the surgery group compared to the control group(P<0.05).Conclusion Surgery mediated systemic inflammatory response is associated with the NLRP3 inflammasome pathway.
