摘  要:糖胺聚糖是一种重要的药用活性寡糖,由于蜗牛糖胺聚糖的提取工艺尚不发达,使得蜗牛糖胺聚糖提取物中具有药理活性的有效结构仍不清楚,本文通过优化糖胺聚糖提取工艺,利用化学酶法联合丙酮循环脱脂工艺,经乙醇沉淀从蜗牛不同组织中提取糖胺聚糖,通过咔唑反应对蜗牛肌肉、内脏及黏液中的糖胺聚糖进行定量分析,结果显示,蜗牛肌肉、内脏及黏液中的糖胺聚糖提取率分别为0􀆰776%,1.961%和0.573%,二糖组分分析显示,蜗牛二糖组成以ΔUA2S-GlcNAc为主(98%),并有少量ΔUA2S-GlcNS存在。生物活性分析表明,从蜗牛肌肉、内脏和黏液中提取的糖胺聚糖均具有较高的抗氧化活性。其中,10 mg·mL^(-1)蜗牛黏液糖胺聚糖的总抗氧化能力(T-AOC)最高,为0.6064μmol·mL^(-1),其DPPH·自由基清除率也最高,为64.15%。本研究为蜗牛来源药用糖胺聚糖活性寡糖的制备分析提供研究基础。Glycosaminoglycan is important polysaccharides with various biological activities which could be used as pharmaceutical ingredient for treatment of some disease.Glycosaminoglycans extraction technology is complex and not developed which limited their application.Till not,snail derived glycosaminoglycans have been little reported making the pharmacological activities of their effective structure still unclear.In this study,Glycosaminoglycans were extracted from snail muscle,snail visceral and snail mucus,using chemical enzymatic degrading method.Acetone was used for sample dehydration while ethanol precipitation was uesd for final extraction.Our results showed that the extraction rates of Glycosaminoglycan in muscle,viscera and mucus were 0.776%,1􀆰961%and 0.573%,respectively.Disaccharide composition analysis of glycosaminoglycans showed that the main disaccharide unit wasΔUA2S-GlcNAc(98%)with a small amount ofΔUA2S-GlcNS was observed.Bioactivity analysis revealed that glycosaminoglycans extracted from snail muscle,visceral and mucus both showed high antioxidant activity.Among the extracted samples,10 mg·mL^(-1)of snail mucus glycosaminoglycan revealed the highest total antioxidant capacity(T-AOC)which is 0􀆰6064μmol·mL^(-1),and the the highest DPPH·radical scavenging rate which is 64.15%.This study provided basis for the preparation and analysis of the active oligosaccharides of snail-derived medical glycosaminoglycan.
