摘 要:游离巯基很容易连接形成二硫键,对角蛋白溶液的稳定性及再生蛋白韧性都会产生不良影响,因此对巯基进行适当保护有助于提升角蛋白的性能。为此,文中分别选择聚乙二醇(PEG)、聚乙烯醇(PVA)及十二烷基硫酸钠(SDS)3种物质分析其对巯基的保护效果。以保护效果最好的PEG为例,分析了其相对分子质量和浓度对巯基保护效果的影响,同时采用动态光散射、荧光光谱及拉曼光谱对其胶束粒径、疏水氨基酸暴露程度和角蛋白二级构象等进行了测试表征。结果显示,PEG可以通过疏水作用力和氢键等方式与角蛋白分子间形成高分子复合物,PEG相对分子质量越大、浓度越高,所形成的复合物粒径就越大,保护效果也越好;而且,复合物能使角蛋白分子舒展,减弱了巯基无序交联形成二硫键的比例,在固化后的角蛋白混合膜中,二级结构变得规整有序,膜的柔韧性和透光性均显著增加。Free thiol groups are easily connected to form disulfide bonds,which will adversely affect the stability of keratin solution and properties of regenerated keratin.Therefore,the proper protection of thiol group can help improve the performance of keratin.In this paper,polyethylene glycol(PEG),polyvinyl alcohol(PVA)and sodium dodecyl sulfate(SDS)were employed as main compounds and their protective effects on thiol group were detailed studied.Then,PEG with the best protection effect was chosen as main study object.The influence of PEG molecular weight and concentration on the protection effect of sulfhydryl group was further analyzed.The micelle particle size,hydrophobic amino acid exposure ratio,and secondary conformation were also characterized by dynamic light scattering(DLS),fluorescence spectroscopy,and Raman spectroscopy.The results show that PEG can form polymer complexes with keratin molecules through hydrophobic interaction and hydrogen bonding.Moreover,the Ker/PEG complex can make the keratin molecules stretch and weaken the disordered cross-linking of thiol group.Compared with the keratin film,the secondary structure of PEG/Ker becomes much more regular,and the flexibility and light transmittance of the membrane are also significantly increased.
