摘 要:目的 :探索微小核糖核酸148a(miR-148a)对食管鳞癌干细胞迁移与侵袭能力的影响。方法 :收集新疆医科大学附属肿瘤医院20例食管鳞癌手术标本,PCR法检测癌及癌旁组织中miR-148a的表达。采用流式细胞技术对食管鳞癌细胞系109(Ea109)细胞进行分选,分离侧群细胞(SP细胞)和非SP细胞,PCR法检测miR-148a的表达。慢病毒转染构建细胞SP亚群miR-148a表达差异模型:miR-148a模拟物、miR-148a模拟物对照、miR-148a抑制物、miR-148a抑制物对照,采用划痕实验和细胞迁移实验检测迁移及侵袭能力的差异。结果 :食管鳞癌组织中及Eca109细胞SP亚群中miR-148a表达明显下调,miR-148a模拟物组的细胞迁移及侵袭能力明显下降,miR-148a抑制物组的迁移和侵袭能力最强。结论 :miR-148a的表达可以影响食管鳞癌干细胞的迁移及侵袭能力,miR-148a可能是一种潜在的食管鳞癌治疗靶点。Objective:To explore whether microRNA-148a(miR-148a)affects the migration and invasion of esophageal carcinoma stem cell(ECSC).Methods:Twenty tumor specimens were obtained from patients diagnosed with esophageal carcinoma and underwent radical resection in the Affiliated Tumor Hospital of Xinjiang Medical University.The cancer stem cell-like side population(SP)cells and non-SP cells were harvested from human esophageal squamous carcinoma cells(Eca109)based on a flow cytometry technique.The expression of miR-148a were detected by reverse transcription-quantitative PCR.After lentiviral vectors containing miR-148a mimics,inhibitors and normal controls had been transfected respectively into SP cells,the migration and invasion were detected using a transwell assay.Results:The expression of miR-148a was statistically down-regulated in carcinoma tissue compared to that in paracancerous normal tissue.And,miR-148a showed significant downregulation in SP cells compared to non-SP cells.Transfection of miR-148a mimics resulted in lesser migrated and invasive SP cells,whereas SP cells transfected with miR-148a inhibitors revealed excessive migration and invasion.Conclusions:These findings suggested that miR-148a can suppress migration and invasion of esophageal squamous carcinoma stem cells.And,miR-148a may hold potential as therapeutic tar get of esophageal squamous carcinoma.
