摘  要:为了探究生物信息学软件识别禽白血病病毒E(Avian leukosis virus subgroup E, ALVE)基因插入位点的准确性,试验采用obsERVer流程和位点特异性PCR方法,测定了3只BWEL-SPF鸡全基因组测序数据中的ALVE基因插入位点,并验证了obsERVer流程预测的准确性。结果表明:在3只BWEL-SPF鸡中共识别到4个ALVE基因插入位点,分别为ALVE1、ALVE3、ALVE9和ALVE15;插入位点上下游序列存在规律性,是由6个碱基的基序构成的重复序列;并且这4个插入位点均可以通过位点特异性PCR方法验证,吻合率为100%。说明obsERVer流程可以准确预测测序数据中的ALVE基因插入位点,在BWEL-SPF鸡中存在多个ALVE基因插入位点。To verify the accuracy of bioinformatic methods identifying Avian leukosis virus subgroup E(ALVE)insertion sites,the obsERVer pipeline and locus-specific PCR were used to determine the insertion sites of ALVE gene in the whole genome sequencing data of three BWEL-SPF chickens,and the accuracy of the prediction of obsERVer process was verified.The results showed that there were four ALVE insertion sites identified in BWEL-SPF chickens.The ALVE insertion sites were ALVE1,ALVE3,ALVE9 and ALVE15.The sequence upstream and downstream of the insertion site was regular,which was a repetitive sequence composed of 6 bases.These ALVE insertion sites could be verified by locus-specific PCR,and the coincidence rate was 100%.These results indicated that the obsERVer process could accurately predict the insertion sites of ALVE gene in sequencing data,and there were various insertion sites of ALVE gene in BWEL-SPF chickens.
