摘  要:目的建立青年、老年两个年龄组大鼠神经功能损伤程度相同的大脑中动脉阻塞(middle cerebral artery occlusion,MCAO)模型,以避免因动物模型偏倚引起的脑缺血药物疗效及毒理学机理等研究的实验结果不准确。方法采用雄性SD大鼠,首先通过实验探索适合老年大鼠的线栓型号后,大鼠被随机分为4组:青年假手术组(n=20),老年假手术组(n=20),青年MCAO组(n=20),老年MCAO组(n=20)。建立青年MCAO组、老年MCAO组大鼠神经功能损伤程度相同的MCAO模型。通过改良的神经功能评分(mNSS)、角测试评估、梗死区细胞凋亡情况评估大鼠脑损伤情况。结果青年大鼠选用线栓型号为硅胶头直径0.32 mm、插入深度(18±2)mm,老年大鼠线栓型号为硅胶头直径0.43 mm,插入深度(22±2)mm。通过调整线栓位置,当青年大鼠局部脑血流量(regional cerebral blood flow,rCBF)为基线血流的15%、老年大鼠rCBF为基线血流的25%时,二者术后3 d内神经功能损伤水平差异无统计学意义(P>0.05),梗死区域细胞凋亡程度相同(P>0.05)。结论通过调节大鼠rCBF能控制脑损伤程度,从而可建立青年、老年神经功能损伤程度相同的大鼠MCAO模型,用于脑缺血的相关研究,以避免因动物模型偏倚引起的脑缺血药物疗效及毒理学机理等研究的实验结果不准确。Objective Establishment of a middle cerebral artery occlusion(middle cerebral artery occlusion,MCAO)model in both young and aged rats with equivalent levels of neurological impairment,in order to avoid inaccurate experimental results of drug effi⁃cacy and toxicology caused by bias in animal models of cerebral ischemia.Methods Male Sprage-Dawley rats were used in this study.After exploring the suitable suture for aged rats,the rats were randomly divided into 4 groups:Young sham group(n=20),Aged sham group(n=20),Young MCAO group(n=20),and Aged MCAO group(n=20).The MCAO model with the same degree of neurologi⁃cal impairment was established in the Young MCAO group and the Aged MCAO group.Brain injury was evaluated by modified neuro⁃logical function score(mNSS),Angle test and assessment of apoptosis in the infarct area.Results In young rats,the diameter of the sili⁃cone tip was 0.32 mm and the insertion depth was(18±2)mm.In aged rats,the diameter of the silicone tip was 0.43 mm and the inser⁃tion depth was(22±2)mm.By adjusting the suture position,when local cerebral blood flow(rCBF)was 15%of the baseline blood flow in young rats and 25%of the baseline blood flow in aged rats,there was no significant difference in the level of neurological injury within 3 days after operation(P>0.05),and the degree of apoptosis in the infarcted area was equal(P>0.05).Conclusion By regu⁃lating rat rCBF,it is possible to control the degree of brain damage and establish a rat MCAO model with the same level of neurological function damage in young and old rats,which can be used for related research on cerebral ischemia.This helps to avoid inaccurate ex⁃perimental results of drug efficacy and toxicology mechanisms caused by bias in animal models of cerebral ischemia.
