荧光定量PCR法与常规细菌鉴定法在肠道致病菌检测中的应用效果

摘　　要：目的 探究在肠道致病菌检测中,应用荧光定量PCR法与常规细菌鉴定法的临床效果,并对之展开分析和探讨。方法 本次研究将时间定为2019年1月1日-2021年12月31日,纳入研究的参与对象为:上述时间段内,某医院收集的腹泻患者1286例资料。所有参与患者都进行了粪便采集和检测,检测方法为荧光定量PCR法和常规细菌鉴定法。对检测结果实施详细记录与分析。结果 所有粪便标本中,应用常规细菌检测出沙门菌21例,阳性率为1.63%,志贺菌50例,阳性率为3.88%;应用荧光定量PCR检测的沙门菌与志贺菌病例分别为23例、53例,阳性率分别为1.79%、4.11%。且将荧光定量PCR检测出的感染患者,应用常规细菌再次检测,检测结果完全相符,阳性率为100.00%。结论 荧光定量PCR检测法具有独特优势,不仅能够直接将粪便中的DNA快速提取,对致病菌进行准确判断,同时操作快速、简单,是一种可大范围推广应用的检测方式。Objective To explore clinical effect of fluorescent quantitative PCR and routine bacterial identification methods in detection of intestinal pathogenic bacteria,and carry on analysis and exploration.Methods The study was conducted from January 1,2019 to December 31,2021,participants in study were:data of 1286 diarrhea patients in a hospital during above period.The paper collected stool of all participating patients and carried on test with fluorescent quantitative PCR and routine bacterial identification methods.Test results were recorded and analyzed in detail.Results Among all fecal samples,21 cases of Salmonella were detected by routine bacteria,with positive rate of(1.63%),and 50 cases of Shigella were detected by routine bacteria,with positive rate of(3.88%).(23)and(53)cases Salmonella and Shigella were detected by fluorescence quantitative PCR,with positive rates of(1.79%)and(4.11%)respectively.Infected patients detected by fluorescence quantitative PCR were retested with routine bacteria,detection results were completely consistent,with positive rate of 100.00%.Conclusion:Fluorescence quantitative PCR method has unique advantages,can extract DNA from feces directly and determine pathogenic bacteria accurately,with fast and easy operation,which can be promoted and applied widely.
