摘  要:目的探讨miR-627-3p靶向调控CK2β对上皮性卵巢癌细胞增殖、凋亡、侵袭的影响及潜在的作用机制。方法采用qRT-PCR和Western blot检测正常卵巢上皮细胞HOSE和卵巢癌细胞株SKOV3、ES-2及OVCAR3中miR-627-3p和CK2β的表达;采用双荧光素报告基因实验验证miR-627-3p和CK2β之间的靶向关系;建立miR-627-3p过表达及miR-627-3p和pcDNA-CK2β共转染的SKOV3细胞株,采用CCK-8法检测细胞活性,采用流式细胞术检测细胞凋亡率,采用Transwell法检测细胞侵袭能力。结果HOSE组、SKOV3组、ES-2组及OVCAR3组miR-627-3p的表达水平分别为(1.00±0.12)、(0.21±0.04)、(0.41±0.07)及(0.32±0.06),差异有统计学意义(F=183.037,P<0.05)。与正常卵巢上皮细胞HOSE相比,卵巢癌细胞株SKOV3、ES-2及OVCAR3中miR-627-3p表达显著降低,CK2β表达显著增加,差异均有统计学意义(均P<0.05);CK2β是miR-627-3p的靶基因,且miR-627-3p负性调节CK2β表达。过表达miR-627-3p可抑制SKOV3细胞活性、促进细胞凋亡,抑制细胞侵袭(均P<0.05);CK2β过表达可部分逆转miR-627-3p过表达对SKOV3的生物学影响。结论miR-627-3p靶向调控CK2β进而影响上皮性卵巢癌细胞增殖、凋亡及侵袭。Objective To explore the effect of miR-627-3p targeting regulating CK2βon proliferation,apoptosis,and invasion of epithelial ovarian cancer cells and the potential mechanism.Methods qRT-PCR and Western blot were used to detect the expressions of miR-627-3p and CK2βin normal ovarian epithelial cells HOSE and ovarian cancer cell lines SKOV3,ES-2,and OVCAR3;dual fluorescein reporter gene experiment was used to verify the targeting relationship between miR-627-3p and CK2β;miR-627-3p overexpression and miR-627-3p and pcDNA-CK2βco-transfected SKOV3 cell line was established,CCK-8 assay was used to detect cell viability,flow cytometry was used to detect cell apoptosis rate,Transwell assay was used to detect cell invasion.Results The expression levels of miR-627-3p in HOSE group,SKOV3 group,ES-2 group,and OVCAR3 group were(1.00±0.12),(0.21±0.04),(0.41±0.07),and(0.32±0.06),respectively,there was significant difference(P<0.05).Compared with normal ovarian epithelial cells HOSE,the expressions of miR-627-3p in SKOV3,ES-2 and OVCAR3 ovarian cancer cell lines significantly decreased,and the expressions of CK2βsignificantly increased.The differences were statistically significant(P<0.05);CK2βwas the target gene of miR-627-3p,and miR-627-3p negatively regulated CK2βexpression.Overexpression of miR-627-3p inhibited SKOV3 cell activity,promoted cell apoptosis,and inhibited cell invasion,the differences were statistically significant(P<0.05);overexpression of CK2βpartially reversed the biological impact of overexpression of miR-627-3p on SKOV3.Conclusion miR-627-3p targeting regulating CK2βcan affect proliferation,apoptosis,and invasion of epithelial ovarian cancer cells.
