摘 要:目的探讨马铃薯抗性淀粉(resistant starch,RS)对高脂血症大鼠胰腺组织的病理学及其相关信号通路的作用。方法48只雄性SD大鼠按体质量随机分为空白对照组和模型组,模型组大鼠在建模后,按总胆固醇(total cholesterol,TC)随机分为高脂模型组、阳性对照组、马铃薯RS高、中、低剂量组。干预6周后,镜下观察胰腺组织病理切片;实时荧光逆转录定量聚合酶链反应(real-time reverse transcription quantitative polymerase chain reaction,RT-qPCR)检测胰腺组织中AMP依赖的蛋白激酶(adenosine monophosphate-activated protein kinase,AMPK)、固醇调节元件结合蛋白1c(sterol regulatory element-binding protein 1c,SREBP-1c)、脂肪酸合成酶(fatty acid synthetase,FAS)、固醇调节元件结合蛋白裂解激活蛋白(sterol regulatory element binding protein cleavage activating protein,SCAP)、过氧化物酶体增殖物激活受体α(peroxisome proliferator-activated receptor alpha,PPARα)的信使核糖核酸(messenger ribose nucleic acid,mRNA)表达。结果与空白对照组比较,一定剂量的马铃薯RS可以减轻高脂血症大鼠胰腺病理损伤,尤其是腺泡细胞坏死现象。高脂模型组大鼠AMPK mRNA、PPARαmRNA表达水平降低(P均<0.01),经马铃薯RS干预后,与高脂模型组比较,马铃薯RS高、中剂量组AMPK mRNA、PPARαmRNA表达均增强(P均<0.05)。高脂模型组FAS mRNA、SCAP mRNA、SREBP-1c mRNA表达升高,经马铃薯RS干预后,与高脂模型组比较,马铃薯高、中、低剂量组SCAP mRNA、SREBP-1c mRNA和高剂量组FAS mRNA的表达降低(P均<0.05)。结论马铃薯RS可以通过调控相关信号转导通路表达,改善高脂血症大鼠胰腺组织损伤。Objective To investigate the effects of potato resistant starch(RS)on pathology and its related signaling pathways in pancreatic tissue in hyperlipidemia rats.Methods A total of 48 male SD rats were randomly divided into blank control group and model group according to body mass index.After modeling,the rats in model group were randomly divided into high-fat model group,positive control group and high,medium,low dose potato RS groups according to total cholesterol(TC).After 6 weeks of intervention,pathological sections of pancreas were observed under microscope.The expression of messenger ribose nucleic acid(mRNA)of adenosine monophosphate-activated protein kinase(AMPK),sterol regulatory element-binding protein 1c(SREBP-1c),fatty acid synthetase(FAS),sterol regulatory element binding protein cleavage activating protein(SCAP)and peroxisome proliferator-activated receptor alpha(PPARα)in pancreatic tissues were detected by real-time reverse transcription quantitative polymerase chain reaction(RT-qPCR).Results Compared with the blank control group,a certain dose of potato RS could reduce the pathological injury of pancreas,especially the necrosis of acinar cells in hyperlipidemia rats.The expression levels of AMPK mRNA and PPARαmRNA in the high-fat model group decreased(all P<0.01).After potato RS intervention,the expression levels of AMPK mRNA and PPARαmRNA in high-dose,medium-dose potato RS groups increased compared with those in the high-fat model group(all P<0.05).The expressions of FAS mRNA,SCAP mRNA and SREBP-1c mRNA in the high-fat model group increased,and after potato RS intervention,the expressions of SCAP mRNA,SREGBP-1c mRNA and FAS mRNA in potato high-dose,medium-dose and low-dose potato RS groups decreased compared with high-fat model group(all P<0.05).Conclusion Potato RS can ameliorate pancreatic tissue injury in hyperlipidemia rats by regulating the expression of related signal transduction pathways.
