摘 要:目的:基于IL-6/信号转导及转录激活因子3(STAT3)通路观察维生素D3(VitD3)对子宫肌瘤细胞的影响。方法:选取子宫肌瘤患者的子宫肌瘤组织进行细胞原代培养,采用CCK-8法检测不同剂量VitD3对子宫肌瘤细胞活力的影响,将细胞随机分为3组,采用无显著毒性的剂量0、20、50 U/g进行后续实验。采用CCK-8法检测VitD3对人子宫肌瘤细胞增殖能力的影响;采用流式细胞术检测细胞凋亡;流式分选检测细胞线粒体膜电位的变化;RT-PCR法检测细胞凋亡相关基因Bcl-2、Caspase-9、Bax的m RNA表达水平;Western blot法检测P-STAT3、IL-6及STAT3的蛋白表达水平。结果:与VitD30 U/g组比较,VitD320、50 U/g组细胞增殖率显著降低(P<0.05),凋亡率显著增加(P<0.05),线粒体膜电位明显降低(P<0.05),抗凋亡相关基因Bcl-2的m RNA表达水平显著下调(P<0.05),促凋亡相关基因Bax和Caspase-9的m RNA表达显著上调(P<0.05),并下调p-STAT3和IL-6的蛋白表达水平(P<0.05),具有浓度依赖性。结论:VitD3可能通过介导IL-6/STAT3通路,减弱子宫肌瘤细胞增殖能力并且促进细胞凋亡。Objective:Effects of vitamin D3(VitD3)on uterine leiomyoma cells were observed based on the signal pathway of IL-6/signal transduction and transcription activator of transcription 3(STAT3).Methods:The uterine fibroid tissues of patients with uterine fibroids were selected for cell primary culture,and the effects of different doses of VitD3 on the viability of uterine fibroid cells were detected by CCK-8 method.The cells were randomly divided into three groups,and the doses of 0,20 and 50 U/g without significant toxicity were used for subsequent experiments.CCK-8 assay was carried out to detect the effect of VitD3 on the proliferation of human uterine fibroid cells,cell apoptosis was detected by flow cytometry,the changes of mitochondrial membrane potential were detected by flow cytometry,the mRNA expression levels of apoptosis related genes Bcl-2,Caspase-9 and Bax were detected by RTPCR,the protein expression levels of p-STAT3,IL-6 and STAT3 were detected by Western blot.Results:Compared with VitD30 U/g group,the proliferation rate of cells in VitD320 and 50 U/g groups were significantly decreased(P<0.05),the apoptosis rate was significantly increased(P<0.05),the mitochondrial membrane potential decreased significantly(P<0.05),the mRNA expression level of anti-apoptotic gene Bcl-2 was significantly down-regulated(P<0.05),the mRNA expressions of pro-apoptotic genes Bax and Caspase-9 were significantly up-regulated(P<0.05),and down-regulated the protein expression levels of p-STAT3 and IL-6(P<0.05)with concentration dependence.Conclusion:VitD3 can inhibit and induce apoptosis of uterine leiomyoma cells in vitro,which may be related to the inhibition of IL-6/STAT3 signaling pathway.
